Objective: This study was to observe the hepatic arterial infusion β-elemene emulsion on rabbit VX2 liver transplant cancer intervention effects of β-elemene emulsion after transcatheter arterial infusion in rabbits and its pharmacokinetics process in vivo; and observe β-elemene emulsion infusion treatment of VX2 carcinoma transplanted liver pathology and for bax, bcl-2 (bcl-2 regulation of apoptosis proteins, blocking apoptosis and prolong cell survival time , bax genes that regulate apoptosis, antagonistic role of bcl-2) expression, thus further explore β-elemene anti-tumor mechanism, looking for a better theoretical basis for clinical application. Methods: 42 Japanese rabbits successfully established liver VX2 liver tumor model. Were randomly divided into control group: (A group): 6, 5 hours uniform injection of 50mg blank milk; treatment group (B): 30, 5 hours uniform injection 50mgβ-Elemene which 24 reserved for pharmacokinetic science, 6 and A, C group with pathological and immunohistochemical; C Group: 6, 5 hours uniform injection of 5-FU (5 - FU) 50mg. Through the femoral artery catheter, catheter tip placed in rabbit hepatic artery, by different groups for the corresponding drug infusion. Intervention group B were taken after 24 arterial and venous blood and tumor (tumor made of pulp), which measured drug concentrations. The remaining rabbits in group B and A, C in the intervention group rabbits were sacrificed after 2 weeks, take all the tumor tissue, in the light microscope pathological changes in tumor tissue, immunohistochemistry bax and bcl-2, evaluate different options treatment. Results: 1 Gross specimen: (a) three groups were seen in tumor weight difference is large, blank breast tumor weight maximum Elemene infusion group and 5-Fu tumor weight little difference. (2) varying degrees of necrosis: the treatment group showed a homogeneous specimen surface view fish-shaped, cut view of the central region showed different degrees of tumor necrosis, manifested as sallow tofu-like changes, accompanied by a small amount of bleeding, adjacent liver tissue swelling, film tension; blank milk group specimens more bleeding, necrosis light, as small pieces and survival of tumor necrosis interval exists, until the tumor margin, no significant changes in adjacent liver tissue. 2 light microscope: the treatment group showed a tumor coagulation necrosis, manifested as a finely granular unstructured like substance, necrotic tissue completely disintegrated. Tumor liver tissues showed obvious hydropic degeneration. Showed significantly enlarged liver cells, the cytoplasm loose, translucent, but still polygonal cell morphology; tumor and surrounding periportal intimal thickening combined thrombosis. Specimens were seen in all treatment groups with inflammatory cell infiltration. Including 5-FU group showed moderate necrosis, elemene emulsion infusion group showed severe necrosis. 3 Immunohistochemistry: Elemene infusion tumor cells bcl-2 expression was significantly reduced, while the expression of bax greatly increased. Β-elemene group and control group apoptosis-related gene bcl-2, bax protein expression in primary located in the cell cytoplasm and some interstitial tissue, while a small number of visible cytoplasm and membrane coloring, was spotty or patchy distribution, a small amount of normal liver tissue expression. bcl-2 expression rate of β-elemene group was 10.09 ± 1.23%, 5-FU group was 30.25 ± 4.29%, while the empty milk group was 51.58 ± 12.40%, pairwise comparisons differences were statistically significant (p lt ; 0.05); bax expression rate of β-elemene group and 5-FU group and blank milk group were 75.29 ± 17.16% and 41.90 ± 9.39% and 18.68 ± 6.44%, pairwise comparisons differences were statistically significant (p lt; 0.05). β-elemene group and 5-FU group and blank group bcl-2/bax milk ratios were 0.15,0.73,2.85, pairwise comparisons were statistically significant differences (p lt; 0.05). 4 Pharmacokinetics: After arterial blood group was higher than that taken 2 hours after reperfusion after taking group, the difference was statistically significant (p lt; 0.05), arterial blood is taken after the group was higher than taken after 2 hours after reperfusion group, the difference was statistically significant (p lt; 0.05), tumor pulp slurry that is to take the average group tumor drug concentrations higher than two hours after taking group, but the difference was not statistically significant (p gt ; 0.05). Conclusion: β-elemene arterial infusion can be effectively treated rabbit VX2 liver transplant cancer. In clinical work, the use of β-elemene continuous infusion therapy for liver cancer is the alternative method.
