One of the multiple tumor nasopharyngeal carcinoma (Nasopharyngeal carcinoma, NPC) for China, the incidence rate of first ENT malignant. Nasopharyngeal cancer occurred in the nasopharynx pharyngeal recess, the top of the rear wall, followed by the side wall, rarely occur in the anterior wall and the bottom wall. Due to the the nasopharynx hidden anatomical location, not the typical early symptoms of nasopharyngeal carcinoma, can be expressed as aldicarb in blood; unilateral tinnitus, ear occlusion flu and headaches. Including a large number of studies have demonstrated that: in the presence of a large number of differentially expressed genes in the carcinogenesis process of NPC involving gene signal and protein delivery oncogene original oncogene, immune-related genes, apoptosis genes, and DNA-binding transcription and transcription factor range . Currently considered nasopharyngeal carcinoma related to genetic factors, viral infections and environmental factors. These factors will ultimately have on gene and protein expression levels. In recent years, research has shown that cyclooxygenase-2 (cyclooxygenase, COX-2) and epidermal growth factor receptor (epidemal growth factor receptor, EGFR) is not only involved in cell proliferation and differentiation, but also in tumor invasion and metastasis and play an important role in the process of angiogenesis, promoting tumor occurrence and development. However, few studies in the NPC, and the results are inconsistent. Ki67 is a proliferating cell nuclear antigen, a cycle-related genes, its expression and tumorigenesis, development, invasion, stage, metastasis, and studies have shown that the prognosis and to determine its index level can be used to assess the malignancy the risk of distant metastasis. The p53 gene is a tumor suppressor gene, is a major negative regulator of cell proliferation, and to maintain the normal function of the cells through regulation of cell cycle and initiate apoptosis. Objective: To study nasopharyngeal carcinoma, COX-2, EGFR, Ki67 and p53 protein expression, and analysis of the correlation between them, in order to be helpful to the diagnosis, treatment and prognosis of nasopharyngeal carcinoma. MATERIALS AND METHODS: Collect the Second Affiliated Hospital of Dalian Medical University between 2002 and 2007 archive nasopharyngeal paraffin blocks of 44 cases, including 32 cases of cervical lymph node metastases, 12 cases without lymph node metastasis; randomly taken from the same period in the nasopharynx of chronic catarrh biopsy wax block 32 cases for control. HE staining of specimens made of normal tissues, the light microscope, to determine pathological diagnosis again. Using immunohistochemical methods detect tissue COX-2, EGFR, P53 and Ki67 protein expression and analysis of the correlation between them. Final application SPSS11.5 statistical software, experimental results were statistically analyzed by t-test and rank sum test, while the use of Spearman determine the correlation, significantly different standards for P lt; 0.05. Results: 1.COX-2 positive expression in the NPC group average score of 3.9886 ± 2.31900; positive expression rate was 93.2%. Positive expression in chronic catarrh group average score of 5.6250 ± 2.64880; positive expression rate was 100%. COX-2 expression in nasopharyngeal chronic catarrh group higher than the NPC group (P lt; 0.05). 2.EGFR positive expression in the NPC group average score of 3.0682 ± 2.47215; positive expression rate was 79.5%. Positive expression in chronic catarrh group average score of 5.5156 ± 2.16081; positive expression rate was 100%. EGFR expression of the nasopharynx chronic catarrh group higher than the NPC group (P lt; 0.05). 3.Ki67 positive NPC group index average of 25.7841% ± 26.70345%; positive expression rate was 72.7%; high expression rate of 34.1%. In chronic catarrh group positive index average of 5.7656% ± 6.24627%; positive expression rate was 37.5%; high expression rate of 0%. Significant differences between the two groups (P lt; 0.05), the NPC group Ki67 expression than the nasopharynx chronic catarrh group. 4.p53 positive expression in NPC group average score of 3.4886 ± 2.78386; positive expression rate was 84.1%. Positive expression in chronic catarrh group average score of 1.5313 ± 1.29476; positive expression rate was 71.9%. Significant differences between the two groups (P lt; 0.05), the NPC group p53 expression is higher than the the nasopharynx chronic catarrh group. NPC group of COX-2, of EGFR, Ki67 and p53 expression in lymph node metastasis between the groups was no significant difference (P gt; 0.05). 6. COX-2 and EGFR expression in nasopharyngeal carcinoma were positively correlated, the correlation coefficient was 0.349 (P lt; 0.05). Conclusion: 1.COX-2 and EGFR expression rate of nasopharyngeal carcinoma and nasopharyngeal chronic catarrh organization, suggesting that chronic inflammation of the nasopharynx and nasopharyngeal occurrence, development-related, and not appropriate as the nose pharyngeal cancer pathology diagnostic indicators. COX-2 and EGFR expression in nasopharyngeal carcinoma were positively correlated, suggesting that COX-2 and EGFR in nasopharyngeal carcinoma development may influence each other, common or collaborative work. Positive expression rate was significantly higher than in nasopharyngeal carcinoma 3.Ki67 the nasopharynx chronic catarrh group prompted Ki67 as one of the indicators to determine the degree of malignancy and prognosis of nasopharyngeal may determine precancerous populations at high risk individual biomarkers. One of the positive expression rate of nasopharyngeal carcinoma was significantly higher than the nasopharynx of chronic catarrh group 4.p53, suggesting that p53 protein with nasopharyngeal carcinoma, closely related to the development; pathological diagnosis of nasopharyngeal carcinoma and can be used as reference indicators . 5. Nasopharyngeal COX-2, of EGFR, Ki67 and p53 expression is not yet as a useful indicator to determine the presence of lymph node metastasis.
