Abstract

[Objective] atopic dermatitis (Atopic Dermatitis, AD) is a recurrent chronic itching of the skin, and can be associated with serious complications, a large impact on the quality of life of patients, but the treatment is very difficult, so a long time AD has been at home and abroad dermatology research hotspot. Atopic dermatitis is an inflammatory skin a Th1/Th2 cell subsets imbalance allergen in the environment play an important role in the occurrence and development of AD. Fufangfulingancao soup is eczema treatment principles in accordance with Chinese medicine treatment, may have the role of Th2 and Th1 cell subsets Chinese proprietary screening test by animal experiments. In this paper, ovalbumin (OVA) BALB / c mice animal models of atopic dermatitis lesions induced by percutaneous sensitization, further study of a single herb's anti-allergy and the role of cytokines the Fufangfulingancao soup, so as to explore the pathogenesis of AD, to lay the foundation for the development of traditional Chinese medicine treatment of AD. [Method] 170 BALB / c female mice (body weight 25 to 30g) were randomly divided into 17 groups, each group of 10 were cyproheptadine, duckweed, gardenia, Alisma, berberine, cicada, Poria red peony, Nepeta, Cork, licorice, herb, wind, Angelica, double flowers, the saline group and the hydrocortisone group. Electricity razor in the day before the experiment in mice abdomen to the hair, an area of ??about 3 cm2 experiments in solution 20μl coated to give 100mg/ml ovalbumin (OVA) to the hair at the daily, seven days in a row sensitized . Each group of 10 BALB / c mice were early, the late twice daily oral gavage drugs. The second week, the third week of normal animals. Week Four: Repeat the first week of the experimental procedure, and animals were sacrificed 24 hours after induction of the seventh day, blood and a patch of skin specimens. By enzyme-linked immunosorbent assay (ELISA) serum IgE, IL-2, IL-4, IFN-γ level. Sensitized skin tissue block made of conventional HE staining, light microscope, micro-scale mesh eyepiece measured for single blinded count per mm2 of inflammatory cells. [Results] 1. Medicine serum IgE levels in mice atopic dermatitis animal model: compared with the saline group, double flowers serum IgE levels significantly lower statistically significant difference (P ≤ 0.01 or P ≤ 0.001); reduce of Coptis group, cicada group, Poria group, red peony group, windproof group serum IgE levels, statistical differences (P lt; 0.05); 2. serum IgE levels, select Chinese medicine Coptis Chantui , Poria, Chishao, windproof, double flowers detection of serum IL-2, IL-4 and IFN-γ levels. The results showed that compared with the saline group, Chantui group, Poria group, windproof group, double flowers of serum IL-2 levels increased statistical difference (P lt; 0.05); Chantui group, the Poria group, red peony group , wind group, double flowers of serum IL-4 levels lower statistical difference (P lt; 0.05); Poria group, Chishao, group, double flowers of serum IFN-γ levels were significantly increased, statistically significant differences (P ≤ 0.01 or P ≤ 0.001); the Coptis groups, windproof serum IFN-γ levels increased statistically different (P lt; 0.05); Herbs on mouse atopic dermatitis animal model skin the impact of the loss of inflammatory cells: The results showed that, compared with the saline group, the number of inflammatory cells in the the duckweed mice skin lesions significantly reduced, a statistically significant difference (P ≤ 0.01 or P ≤ 0.001); coptis group, the Poria group, Chishao, Group, windproof group the number of inflammatory cells in the, double flower mice lesions decreased statistically different (P lt; 0.05). On this basis, further monitoring mice atopic dermatitis animal model of serum IL-2, IL-4 and IFN-γ levels, the results showed that the cicada, wind, double flowers, Poria can up-regulate IL-2, cicada, Poria, red peony root, red peony, double flowers, wind can inhibit IL-4, berberine, Poria, double flowers, the wind can upregulate IFN-γ, effective mechanism may be adjusted by reducing the levels of IgE as well as participate in Th1/Th2 cells play a role in , IFN-γ can inhibit the increase of Th2 cells, to reduce the formation of IL-4, and further the inhibition of synthesis of IgE related mechanism needs depth. In addition, duckweed, berberine, Poria, TPG, double flowers, wind, when restitution can inhibit skin lesions of inflammatory cells, consider Chinese medicine, anti-inflammatory, anti-allergic effects.
