Topic : Objective : To determine the reliability of dry paper method and wet paper method for screening antifungal activity of traditional Chinese medicine . Method : the most important human pathogenic fungus Candida albicans (Candida albicans) as a biological model activity measurement using dry paper method and the wet paper method of Chinese medicine crude extracts and essential oils against Candida albicans . Method on dry paper method and wet paper method to better meet the formula Z2 = A logQ - M , that is proportional to the logarithm of the square of the diameter of the inhibition zone and pieces of paper containing a dose and dry paper method and no significant difference between the wet paper method . We then measured with a dry paper method and wet paper method of the alcohol extract 27 kinds of traditional Chinese medicine and 29 kinds of plant essential oil extracts activity against Candida albicans , snow tea , heavy floor and cinnamon oil . significant inhibitory effect . Conclusion: wet paper method have not been reported , and for the complex solubility characteristics of crude extracts of traditional Chinese medicine , the wet paper method as long as the right way , can be used as dry paper method complement activity in traditional Chinese medicine to provide more evidence for screening . Topic : Objective: Screening of Saccharomyces cerevisiae use of non-fermentable carbon source gene . Methods: The first 384 -pin photocopying board photocopying Saccharomyces cerevisiae gene deletion mutant library in YPD, YPE glucose , ethanol and glycerol as the carbon source and YPG medium comparative gene deletion strains in complete medium YPD and non-fermentable carbon source culture yl YPE and YPG growth to identify the strain grown in the medium of the non-fermented carbon defects , and then the concentration gradient of these strains with dilution method further verify that they use non - fermentation carbon source of defects , and thereby determining not to use the non-fermented gene -deficient strain of a carbon source . Results: 172 YPE and YPG medium on , have demonstrated growth defect 16 is growing well and grown in YPG defective in YPE 35 is grown in YPG good growth on the YPE defective 's . Conclusion: the discovery of gene function , provide important information to study the metabolism of a carbon source for Saccharomyces cerevisiae clarify Saccharomyces cerevisiae Central Africa fermentation carbon utilization related metabolic mechanism .
