Abstract


The Saccharomyces cerevisiae ScRck2 a Ser / Thr protein kinase , acts on the the HOG pathway downstream Hog1-MAP Kinase phosphorylation and activation . A Hog1 binding loci ( TILQR589R590KKVQ ) and the R589A/R590A pairs of point mutations can not be mediated MAPKK Pbs2pDD lethal effects caused by the existence of the end of the protein C in Rck2 . Rck2p C - terminal 123 amino acid residues containing the kinase activity independent inhibitory sequences , therefore deletion of the 123 amino acid residues may be mediated lethal effects caused by the Pbs2pDD , while missing the segment of the amino acid residues may inhibit the Rck2p The degradation in the case of overexpression . To further determine the kinase activity of the functional areas of self- suppression sequences , we constructed a C-terminal deletions of 27, 60 and 90 amino acid residues mutant ScRck2p △ 27aa ScRck2p △ 60aa and ScRck2p △ 90aa . These mutants were PGAL-Pbs2DD cotransformation to rck2 deficient strain , and found that the cells can not be mediated the MAPKK Pbs2pDD caused lethal effects ScRck2 △ 27aa , the C-terminal 27 amino acid residues do not contain the kinase activity independent inhibitory sequences . The mutant protein overexpression results show that , ScRck2p △ 60aa degradation occurs in the case of overexpression functional region exists in the C-terminal 60 amino acid residues and 123 amino acid residues between control ScRck2 Degradation .
