Recent studies have shown that , The astrocytes can accept external stimuli , and respond to calcium signals in the form , this calcium signal also become a bridge for information exchange between astrocytes and other cells or tissues . These results reveal that astrocytes are important participants of the activity of the central nervous system function . Calcium signal generation mechanism and function is not very clear , an important reason is that no one at the same time with lossless , strength , controllable , and do not need to add calcium signaling induced by exogenous reagents . In this paper, the use of femtosecond pulsed laser stimulation of astrocytes in vitro induced elevated intracellular calcium . Different light intensity of stimulation induced astrocytes produce the two calcium increase mode : S - type calcium elevated or H - type calcium rises . Light stimulation after the entry of exogenous PI dye was observed under the high power lens to elevated extracellular calcium - dependent reversible change of the cell membrane and calcium , confirmed photoluminescence perforation effect femtosecond laser can induce cell calcium increase . Elevated light perforation induced calcium can cause calcium wave different calcium wave caused by the two calcium elevated mode range : S - type calcium increases calcium wave radius of 80.2 ± 7.3 μm , H - type calcium increases The calcium wave Radius 176.6 ± 12.1μm. The Pharmacology evidence both calcium caused by elevated calcium waves are extracellular the ATP signaling factors P2Y receptor mediated . This stimulation method used in mixed culture neurons on astrocytes , elevated S - type calcium and calcium wave does not cause calcium activity of neurons , while the H - type calcium increases and calcium waves can cause nearby neurons synchronous calcium increase . This shows that the intensity of the reaction of astrocytes neuronal calcium activity . These results indicate that the perforated effect of femtosecond pulse laser light can induce the astrocyte calcium increase and calcium waves with lossless controlled intensity does not change the intracellular and extracellular environment , etc. , is a new kind of induced calcium signal.
