Continuous Pretreatments for Improving Saccharification Efficiency of Corn Straw by White Rot Fungi under Non-sterile Conditions

The use of straw for fuel ethanol production , the utilization of waste , can also ease the increasing scarcity of fossil energy crisis . White rot fungi pretreatment straw , can improve the straw saccharification efficiency in fuel ethanol production process , while the white-rot fungi pretreatment , a key issue is the open mode solution . The paper through the screening of commonly used laboratory strains successfully constructed two types of open system : ( 1 ) itself is conducive to the growth in the open on the straw , but saccharification ineffective the BP2 open system ; (2) saccharification , but not 2538 open system conducive to culture in the open on the straw . On this basis , the successful realization of two continuous open mode . Culture conditions single factor test results show that : culture conditions training time and inoculation amount of impact on the system of two open effect is significant , which cultivate extremely significant number of days ; while the moisture content of medium aqueous pH value is not significant with , but two factors saccharification rate the impact there is a favorable range . Optimized by orthogonal test the BP2 open system optimal culture conditions : incubation time of 15 days , the pH value of 7.0 , the inoculation was 38.5% , the substrate water content of 77.8% ; optimal culture conditions of the 2538 Open Architecture : training days to 15 days , the pH value of 3.0 , the inoculation was 42.9% , a water content of 71.4% . Under the best conditions , both saccharification rate were 32.3% and 44.6 % , 26.2% and 69.8% , respectively, higher than the control . Continuous experiment on BP2 and 2538 open system to achieve the initial amplification , BP2 can successfully achieve continuous open three batches , glycosylated improved obviously . 2538 continuous open although the with culture batch increase in glycated effect of variation , but can be successfully achieved in five batches continuous open and saccharification significantly improved compared to the control . The impact of continuous open culture the number of batches a key reason for the increase of bacteria in the system , which affects the white rot fungi in a row to open the dominant niche in the culture system . For the first time successfully constructed the continuous opening of the two types of solid state fermentation of straw pretreatment mode to explore the influence of culture conditions on the open system saccharification optimal culture conditions to optimize the two types of system , the straw into the enlarged opening of the fuel ethanol research pretreatment provides a theoretical and technical support .
