Total glucosides of paeony（TGP） and astragalosides（ASTS） have siginificant anti-inflammatory, anti-oxidative and immunomodulatory effects.SQDG is the effective compound extracted from Radix Paeonia Pall and Radix Astragali by the proportion of 4:1 by uniform design. It contains paeoniflorin（Pae） and astragaloside（astragaloside IV,AST IV） , etc.High pressure liquid chromatography （HPLC） was used to determine the purity. The quality control criteria meeting the requirement of new drug in China has been established.According to our team’s previous researches, liver protective effect of SQDG was studied and compared with TGP and ASTs.It shows that SQDG had stronger antioxidation effect than TGP and ASTs. For further understanding the immunomodulatory effect of SQDG, the author investigated the modulatory effects of SQDG on the humoral immunity and compare the effect with （TGP）, ASTs and TGP plus ASTs （TGP +ASTs）.Aim To investigate the modulatory effects of SQDG on the humoral immunity in normal mice,in immunodepression mice and in immunoenhancement mice,and compare the effects with （TGP）, ASTS and TGP plus ASTs （TGP +ASTs）. Methods Sheep red blood cell（SRBC） model induced by SRBC and cyclophosphamide （Cy） inhibited and potentiated SRBC model were used . SQDG (30,60,120 mg·kg-1) were given intragastrically（ig） once daily for 7 consecutive days. LPS-inducedB thymocytes proliferation was measured by MTT assay. the calculation of 50% hemolysin value （HC50）,formation of IgM antibody in splenic cells was measured by 721 spectrophotometer. Compare the difference of the effect of SQDG,TGP, ASTS and TGP+ASTS on the SRBC model.Results （1） In normal immune state,SQDG(120 mg·kg-1) could increase indexes of thymus and spleen,HC50, IgM,B thymocytes proliferation. SQDG(30,60,120 mg·kg-1) could increase and restore all indexes in mice immunosuppression model caused by Cy(150 mg·kg-1). SQDG(30,60,120 mg·kg-1) also could induce and restore all indexes in mice immunoenhancement model caused by Cy(250 mg·kg-1).（2） Comparing with TGP(140mg·kg-1) and TGP+ASTs (105+15mg·kg-1),SQDG (120 mg·kg-1) has superiority in increasing spleen index,HC50,IgM,B thymocytes proliferation in SRBC mice ; SQDG (30,60,120mg·kg-1) was superior to TGP (35,70,140mg·kg-1) and TGP+ASTs (26.25+3.75,52.5+7.5,105+15mg·kg-1) in increasing all indexes in Cy-suppressed SRBC mice; In Cy-potentiated SRBC mice,SQDG(30,60,120mg·kg-1) has obvious superiority in inducing and restoring all indexes compared with TGP(35,70,140mg·kg-1)、TGP+ASTs(26.25+3.75,52.5+7.5,105+15mg·kg-1).Conclusion SQDG possesses a dual modulatory effect on humoral immunity in mice and has obvious superiority compared with TGP and TGP+ASTs of similar doses.
