Abstract


Worldwide blood supply, virus contamination and other issues to promote the study of blood substitutes. Transfusion sources are limited and may cause serious adverse reactions after infusion, and thus domestic and foreign scientists to the oxygen-carrying solution as blood substitute for more than a century of exploration. To date, several first generation of red blood cell substitutes have been expected to be allowed to market. Its unique physiological characteristics of a variety of products, biological activity and side effects. This study, from liposomes hemoglobin (liposome Encapsulated hemoglobin, LEH)'s point of view, research and development as an oxygen therapeutic agent may become the future development direction of development of red blood cell substitutes. About the research: Objective: To determine the effective preparation and identification of microcapsules coated artificial red blood cell hemoglobin and micron. Methods: Preparation of high purity by hypotonic SYSTEM high concentration of membrane protein, and then uniform microcapsules prepared according to the principle of the interfacial polycondensation method, nozzle, containing 2 ,3-DPG, membrane proteins and hemoglobin water phase was added dropwise directly to the phospholipid-containing vegetable oil, by stirring at high speed so that the capsule the heart material (2 ,3-DPG and hemoglobin monomer and dimer built unstable tetramer) is formed around the the single spherical membrane shell-type microcapsules. Results: The microcapsules coated hemoglobin its spherical microcapsules, smooth and uniform surface, the microcapsules of 2-5μm in diameter (electron microscope estimated value). Artificial erythrocytes TEM: single encapsulated particles of spherical shape, smooth surface, some slightly convex surface, the average particle diameter of 3μm, and the minimum particle size of 1μm; Scanning electron microscopy showed that: the high electron density of the grain interior, uneven fibrous and granular staggered distribution for hemoglobin. Embedding of artificial red blood cells was 53%, 40.9% leak rate of 4 ° C saline 15d release. Conclusion: The two-phase inversion emulsification method may be prepared by the micron level artificial erythrocytes, and having the biological activity of the carry and release oxygen and carbon dioxide. A variety of modified hemoglobin fluorocarbon emulsions, hemoglobin, microcapsules rapid development of technology, some of which have formed large-scale preparation process. However, blood substitutes do not have a complete evaluation system, a comprehensive assessment of the blood substitute product is still difficult. Therefore, another important part is to research the blood substitutes based, combined with the latest developments in the world, from the point of view of the evaluation of blood substitutes, to investigate blood substitutes evaluation parameters around the physical and chemical properties of blood substitutes , effectiveness and safety, preservation conditions are described.
