Abstract


Objective: To investigate the MDR1-siRNA and paclitaxel cell lines SKOV3-TR30 - resistant adhesion of human ovarian epithelial cell carcinoma . Methods : 1 . Chemical synthesis contention MDR-1 gene as a target goal siRNA, liposomes law , transfected SKOV3-TR30 cells. 2.EDTA mediated cell separation experiments to test the ability of cells to the the plastic basement membrane adhesion before and after transfection . 3.MTT assay transfected cells and the basement membrane and extracellular matrix components , LN and FN adhesion ability . 4.Western Blot detection of cell surface adhesion protein CD44v6 amount of protein expression changes before and after transfection . Immunofluorescence to observe the changes of cell surface CD44v6 . 6 Flow Cytometry the membrane protein CD44v6 changes do quantitative comparison. Results : 1.EDTA mediated cell separation experiments showed SKOV3-TR30/MDR adhesion of cells to the plastic matrix decreased significantly ( P <0.05 ) . 2 . Experiments show that the adhesion of the extracellular matrix and basement membrane components , the SKOV3-TR30/MDR cell adhesion to LN and FN decreased significantly ( P <0.05 ) than those before transfection . The 3.Western the Bolt Law and immunofluorescence observed before and after transfection adhesion molecule CD44v6 cells , SKOV3 and SKOV3-TR30/MDR cells showed no expression, SKOV3-TR30 and SKOV3 - TR30 / non cell expression , The Flow Cytometry display CD44v6 percentage of positive cells were 30.72% , 31.63% , 66.09 % and 68.15 % , SKOV3-TR30/MDR group was significantly lower . After discussion : MDR1-siRNA liposome-mediated transfection of ovarian cancer, paclitaxel resistant cell lines SKOV3-TR30 , its adhesion capacity decline.
