Abstract


By mRNA differential display (Differential Display Reverse Transcript PCR, referred DDRT-PCR) technology is relatively normal for iron and iron- deficiency stress -efficient wheat ( Beijing -411 ) and iron inefficient type wheat ( three genera Mai -3 ) of differences in gene expression . The results showed that both iron deficiency stress -induced expression of certain genes , can also inhibit the expression of some genes ; iron deficiency stress -efficient varieties ( Beijing -411 ) differences in gene expression than iron inefficient type wheat ( three genera Mai -3 ) significantly , indicating their ability to resist iron Deficiency indeed different. Pick out the use of differential display cDNA fragment as a probe the same from the iron -efficient wheat ( Beijing -411 ) in the normal stress treatment for iron deficiency apical and extracted total RNA by Northern hybridization , proved the difference fragment cDNA2 ( cloned then divided cDNA2-1 and cDNA2-2) and cDNA3 expression is regulated by iron deficiency stress suppressed. Sequencing and sequence analysis showed that , cDNA3 fragment encoding mitochondrial proteins with common wheat , and with the formation of the mitochondrial genes Cytc rpslA 99 % homology , and Arabidopsis plants with several dishes Cytc formation of the mitochondrial genes , 92 % or more homology ; initially identified after analysis of wheat rpslA gene expression in iron-deficiency stress weakened , Cytc formation decreased, affecting respiration and ATP generation . cDNA2-1 fragment with the ATP-binding-transporters have strong family resemblance , cDNA2-2 fragment with dDTP-4, 6 - glucose dehydratase has some homology , suggesting that these two genes in iron-deficiency stress may be weakened expression .
