Abstract


Corn is an important food, feed and cash crops , how to improve corn production , improve the breeding environment , cultivate fine varieties of the main problems faced by the corn breeding . Corn mature embryos as explants build corn regeneration system has also become a new area of ??research recently . Corn mature embryos derived mature embryos as explants callus induction eventually build the process of regeneration system , subject to seasonal restrictions , but because of its specificity , making the training process more difficult. In this paper, the problems encountered in the process of mature corn callus analysis, the results are as follows : 1 . Maize mature embryos most likely to occur in the course of callus induced browning , solve the browning mature embryos induced the most important issue in the process of callus , the test first time screening test observed callus growth of pre-treatment with 2,4-D 24h corn mature embryo explant material for optimum callus organization close texture , root and shoot differentiation phenomenon is relatively weakened, and the browning phenomenon is significantly lower than other treatments . 2 in to filter out the relatively weakest browning culture time after the conditions of the medium filter , add significant control effect of additives such as activated carbon , mannitol , vitamin C and auxiliary to the growth regulating agent 2 browning , 4-D ,6 - BA , the study found that adjusting the mannitol added to the medium for 20 mg / L with 2,4-D , 6 - BA , the concentration as 2,4 - D4mg / L ,6 - BA0 .2 mg / L browning the lowest rate , significant difference . Study also found that 2,4-D corn callus Following the significant effect of culture and differentiation of cultured maize callus from mature embryos subculture , when the concentration of 2,4-D to 1.5 mg / L ,6 - BA concentration for the 0.2 mg / L , the growth of callus in good condition ; 4 in corn callus differentiation culture , when the concentration of 2,4-D is the concentration of 2.0mg / L ,6-BA 0.2mg / L , the the callus differentiation ability .
