Abstract


Micro - column liquid chromatography analytical instruments miniaturization research directions , and it has a high separation efficiency, low consumption and pollution , and other advantages . In this paper, the existing instruments and components packed capillary liquid chromatography system , carried out the study of a wide bore the filled capillary liquid column in the chromatography system . The stigma of a zero- dead volume / column tail structure and a homogenate filler material storage pool evenly into the column tube to study the preparation of internal diameter 0.5-1.0mm stainless steel wide caliber filled capillary LC columns . Examine the direction of the filling system , of pressurized solvent homogenate solvent , homogenate concentration and filling pressures of different factors such as the column efficiency . Equivalent flow rate / equivalent board relations and asymmetry factor column performance evaluation results show that the method of preparation of the column column efficiency more than 70% of the theoretical value repeatability of 6% RSD, stability is also very good . For the analysis of real samples with satisfactory results . Wide caliber packed capillary liquid chromatography column filled capillary liquid chromatography gradient elution , and from the retention time , peak height and peak area gradient elution device evaluation . The experiments show that : the wide caliber packed capillary liquid chromatography can use existing liquid chromatography system eluting devices , chromatography analysis , and promote the practical use of the micro-column liquid chromatography .
