Abstract

Spot Mao (Erianthus arundinaceum) sugar cane (Saccharum L.) is one of the closely related species, with a variety of excellent resistance to sugarcane breeding germplasm. This election sugarcane relatives are the plant spot Mao study material by RT-PCR and RACE clone glutamate pathway in the process of proline metabolism and ornithine way a key enzyme gene delta-OAT and P5CS. Using real-time quantitative PCR on PEG, NaCl and ABA deal with two different time gene expression analysis, to explore the of two biosynthetic pathway in the process of accumulation of proline primary and secondary relationship, and trying to understand the spot under different processing conditions Mao prolyl acid metabolism and drought resistance. Isolated from the spot Mao encoding ornithine-delta-aminotransferase gene (Ea-delta-OAT) full-length cDNA sequence of 1720bp, encoding 454 amino acids, GenBank accession number (EU113256). With mammals, higher plants, microorganisms delta-OAT gene encoding the amino acid sequence homology comparison, spot Mao EA-delta-OA Jin with the closely related genus sugar cane highest homology (87%) , while the second highest homology with other higher plants (approximately 70%), the lowest homology of the same animal (approximately 60%). Spot Mao Ea-delta-OAT gene encoding the amino acid sequence of the 5 'end was not found mitochondrial targeting sequence, with sugar cane Sc-δ-OAT gene. The spot Mao Ea-δ-OAT gene with a complete ornithine aminotransferase Ribbon ROCD the. Obtained the spot Mao △ to 1 - pyrroline - 5 - carboxylic acid synthase gene (EA-P5CS) cDNA full-length 2563bp 5 'end non-coding region of 103bp. Contains a 2148bp encoding 716 amino acid open reading frame (ORF) and 3 'non-coding region 312bp upstream of the poly (A) a polyadenylation base AATAAA, GenBank accession number (EU113257). BLAST analysis of the gene encoding the amino acid sequence in GenBank and its related genera of plant cane homology of 97%, with rice homology of 87%. The P5CS gene encodes a dual function, spot Mao enzyme, N end containing a glutamate kinase (G5K (PROB)), Ribbon, in the C-terminus of a glutamyl semialdehyde dehydrogenase (glutamyl 5-phosphate reductase (G5PR, ProA) function under different stress treatments (PEG, NaCl, and ABA), the Q-PCR analysis of two spots Mao proline metabolism key genes: two genes Q-PCR analysis results are summarized list, and then illustrate the kind of way to play a major role results showed that 30% PEG Stress spot Mao proline accumulation process two metabolic pathways, stress pre glutamate pathway and ornithine pathway while at work, in the late stage of stress Valley histidine way to gradually dominate; the first 6h proline in the Stress 250mMNaCl spot Mao proline metabolic process by glutamate pathways and ornithine pathway co-regulated, followed by ornithine mechanism dominates; 10 mg / proline accumulation spot Mao L ABA treatment process, the combined effect of pre-glutamic acid pathway and ornithine way, ornithine way in the latter part of the dominant spot Mao, there are two genes encoding P5CS, and a for ABA-dependent, is not dependent on the ABA-type EA-P5CS gene cloned into not dependent on ABA type.
