Abstract


The first part of thymic stromal lymphopoietin expression in A549 cells Objective To observe the airway epithelial cell line A549 cells in thymic stromal cells expression of erythropoietin ( TSLP ) and nuclear factor - kappa B ( NF - kappaB ) . A549 cells with the cytokine interleukin- 1 beta ( IL - 1beta ) , IL - 4 or NF - kappa B inhibitor PDTC incubated TSLP mRNA and protein expression was measured using RT - PCR , immunofluorescence method . A549 cytoplasmic expression of TSLP protein , in combination with IL - 1beta IL-4 stimulation 4 h TSLP mRNA expression amount with respect to beta - actin as 0.93 , peaking . With the passage of time , 18 h when the amount of TSLP mRNA expression with respect to beta - actin as 0.31 basic level when reduced to 0 h . PDTC can significantly reduce the peak levels of TSLP mRNA . Conclusion normal airway epithelial A549 cells express TSLP mRNA and protein , of cytokines promote inlet epithelial cells induced expression of TSLP , and by the NF - kappa B signaling pathway regulation . The second part of the drugs on A549 cells thymic stromal cells generate impact of Objective To investigate the the Budi Ned and Astragaloside TSLP expression of A549 cells and NF - kappa B expression . A549 cells and cell factor IL -1beta , IL - 4 or drugs ( Budi Ned Astragaloside ) were incubated , without any cytokines or drugs cultured A549 cells to the control group , the determination of RT-PCR method TSLP mRNA expression , immunofluorescence observed the expression of NF - kappa B ( p65 ) , the Western blot method for detection of cytoplasmic IκBα expression . Results of budesonide significantly inhibited the expression of TSLP mRNA , the obvious inhibition , Astragaloside , ( P gt ; 0.05 ) ; Astragaloside and budesonide were able to significantly inhibit A549 cells , NF - kappa B ( p65 ) nuclear translocation ; Astragalus a glycosides can be significantly increased the A549 Cells IκBα protein levels . Conclusion inhibit TSLP expression and NF - kappaB signaling pathway activation may be an important mechanism of budesonide in the treatment of asthma ; Astragaloside could inhibit NF - kappaB signaling pathway , but did not inhibit the TSLP gene expression .
