Abstract


The 21st century is the era of gene, the completion of the human genome sequencing, functional gene research has also become a hot spot of this century, life science research, this idea also brought tremendous changes to the field of disease animal model. Now we know that all diseases are more or less directly or indirectly linked with the gene. They are divided into three categories: single-gene disorders, polygenic disease and acquired genetic disease. The main principle is established animal models of disease the etiology induction principle. Non-genome era, the traditional establishment of spontaneous and induced animal models of disease, has an important role to play in the research and exploration of the human disease. Today, as people kept informed of the gene, a variety of modern gene knockout technology and transgenic technology, molecular biology technology development, undoubtedly the establishment of disease animal model to produce a revolutionary role, and that role so far already apparent. The combination of modern and traditional methods is the trend for the future establishment of animal models of disease. Human to establish genetic susceptibility to disease animal model, we must first realize the operation of genes. Remove or add a gene transgenic technology are inseparable. Following: microinjection, liposome method, the embryonic stem cells of transgenic technology, electroporation, calcium phosphate precipitation method, and retroviral transfection method used transgenic technology. They have their own advantages and disadvantages. And the sperm carrier method developed in the late 1980s transgenic technology is a bright spot in the field of transgenic technology. And transgenic technology, the species sperm carrier of transgenic technology applications range widely, the integration of the exogenous gene targeted integration by non-homologous recombination, the exogenous gene design less demanding and successful in transgenic article of Transgenic animals generation rate is still relatively high. Sperm carrier of transgenic technology, after all, is still in its infancy, and the understanding of its mechanism is still unclear, and there are many more to be studied and improved. We try to establish the experimental platform sperm carrier of transgenic technology, various steps related factors, the preliminary exploration and analysis, and achieved certain results. There is no doubt that the sperm carrier of transgenic technology as a new force, disease animal model and the study of disease mechanisms will play a decisive role on the function of genes in the overall experimental animals in research.
