Abstract


ObjectiveAlzheimer’ s disease(AD), named senile dementia, is a progressive neurodegenerative disorder with severe cognitive, behavioral, and functional impairment. Among a great deal of healing solution ,locus treatment is an important direction, but at present there are not the safe and effective medicine to resist AD . In the face of a large number of AD patients , It is very urgent and important to invent effective treatments. Traditional Chinese medicine has made great progress, so It is hopeful to use TCM to resist AD. Althouth its etiology is still unknown, it is generally accepted as related to the cholinergic deficit in the brain..In this paper, we determine the effects of Guben Huatan Jiannao Decoction (GHJD) on brain acetylcholinesterase(AchE) and acetylcholine(Ach) levels and behavioral science (Water maze test> Footshock avoidance test) in Alzheimer’ s disease (AD) rats, and analyze the mechanism of action of WDID for AD. MethodsLSD male rats were randomly divided into five groups as the operation group, AD model group, piracetam treated group, high-dose and low-dose of GHJD group.2. We used injection of basal nucleus of Meynert (BNM) with ibotenic acid(IBO) to make AD model. After treating for three weeks, channel water maze task and step-through task were used to observe the ability of learning and memory.3.We used reagent salver to assay the activity of acetylcholinesterase(AchE), and tissue chemistry method was used todetect, the content of acetylcho] ine(Ach) in hippocampus and corpusstriatum areas of brain.Results1. In the behavior experiment, not all the ability of learning gain , the significant differences were observed between control group, piracetam treated group , the high-, low-treated group of GHJD and AD model group(P<0. 01, P<0. 05).2. The activity of AchE was higher in AD model rats than that of control group .piracetam treated , and the high-, low-treated group of GHJD(P<0. 01).3. There was the remarkably improvement on the number of Ach in the three medicine groups(P<0.01 ) , and the activity of Ach in the high-treated group of GHJD was near to the level of control group(P<0. 01) . ConclusionsGuben Huatan Jiannao Decoct ion(GHJD) can improve the disability of learning and memory induced by ibotenic acid , and its effect maybe involve the activity of the Acetylcholine systym.
