Abstract


Lysozyme (lysozyme, LYZ) is a bacterial cell wall hydrolase , direct dissolution of Gram-positive bacteria , Gram-negative bacteria and some fungi have indirect dissolution in clinical medicine and the food industry and other fields has a wide range of applications . The currently using lysozyme primarily egg white lysozyme for the human body has certain immunogenicity and side effects . The natural human lysozyme (hLYZ) from human milk , placenta or saliva to extract a small amount , but sources are extremely limited , unable to meet the medical needs of the clinical . A prokaryotic bacteria such as E. coli expression system expressed at high levels and low cost , but the expression hLYZ inactive inclusion bodies . Transgenic animal bioreactor has a high level of expression and the expression product activity completely , but the relevant technical complexity , has not yet reached the level of development of industrialization . Transgenic chicken oviduct bioreactor technology is more difficult , there is no coverage of high level expression of recombinant proteins . The chicken oviduct transient expression system has low technical difficulty , but existing methods to express the level is low , it is difficult to meet the needs of industrial development . In order to obtain good sensitivity and reproducibility of the quantitative determination of lysozyme , this study reported RBB-R dye-labeled colorimetry and agar diffusion method comparative study , the results show that RBB-R dye-labeled colorimetric the LYZ assay egg white has the advantages of high sensitivity , accuracy and repeatability . Industrial development value rhLYZ the correct sequence hLYZ cDNA was cloned into the tubal specific self - build to expression vector pOV1 and pOV2 the the obtained recombinant expression vectors were named pOV1LYZ and pOV2LYZ . Packet is the recombinant vector and the polyethylene imine (PEI) , were injected with egg-laying hens , egg white lysozyme activity test results showed that the expression level of the pOV2LYZ superior pOV1LYZ , so as expression vectors for further study . 16 laying hens were randomly divided into 2mg / 1 2mg 2 times 3mg / 1 and 4mg / 1 4 test group were injected as a way of PEI coated recombinant vector pOV2LYZ of wing vein . 2mg recombinant vector is determined based on the injection of chicken body temperature and other physiological indices and egg production rate changes and egg white recombinase expression detection
