Surface plasmon resonance (SPR) was used for biosensing thefirst time in 1983 and for imaging applications in 1987 . Withimaging SPR, also referred to as SPR microscopy, new applicationsare possible, e.g. label-free real-time multispot biochemicalanalysis, which can increase sample throughput tremendously. Thepioneering work on imaging SPR was undertaken by Knoll et al., whoinvestigated surface features caused by Langmuir–Blodgett films .They also studied the physical aspects of the technique, includinglateral resolution. The rotating grating coupler setup was firstproposed by this group. SPR imaging is a surface-sensitive opticaltechnique for monitoring the absorption of solution species ontopatterned molecular arrays that have been formed on chemicallymodified metal surfaces in real time. For example, SPR imaging canbe used to study the specific absorption of biopolymers (proteins,antibodies) onto DNA or protein arrays. The array format makes SPRimaging technique a high throughput differential bindingmeasurement that can be used to accurately measure specific organicand biomolecular interactions with a variety of surfaces underidentical solution absorption conditions.The SPR effect is resulted from a charge-density oscillationthat may exist in a glass-Cmetal-Cdielectric stack when theincident p-polarized optical beam undergoes total internalreflection inside a metal-coated glass prism .
