Abstract


Low temperature chilling injury is a serious natural disasters in the agricultural production in the world each year resulting loss of U.S. $ 200 billion . The cold tolerance of crops is controlled by multiple genes , , only clustered resistance gene transcriptional activation in order to effectively improve crop resistance to cold . Cloned and characterized the regulation of the cold functional gene expression of transcription factor become the hotspot of this field of research . The study found that many plants after a certain time cold acclimation , cold resistance will be greatly enhanced . CBF ( C - repeat Binding Factors ) gene in this process , since the role of the adjustment factors . Under low temperature conditions , CBF gene expression activates expression of the COR gene ( Cold -regulated genes ) , thereby enhancing the cold tolerance of a plant body . The cold regulation CBF transcription factor, its function has been demonstrated in a variety of plants , but the regulation mechanisms of the same type of plant . There have been no reported cotton CBF . China is a major cotton producing countries, low temperature chilling injury in cotton-growing areas in northern China , especially Xinjiang cotton area of very serious harm to one of the limiting factors for cotton production . The early hypothermia can lead to delayed sowing ; late low temperature will affect fiber quality . The research cotton CBF genes for enhanced cotton resistance to cold , to extend the growth period , thereby increasing cotton production has a very important significance . In this study, according to the CBF homologous sequences by PCR method was cloned from different cotton varieties and identified a CBF gene in cotton , confirmed the CBF CBF gene specific protein sequence between different cotton varieties CBF gene homology of 97% , 63% DNA homology with the Arabidopsis and tomato homologous DNA 68 % homology with alfalfa 69% , only 40% homology with the barley . CBF family members , 3 to 5 is determined by Southern hybridization of cotton and other cotton CBF genes for cotton BAC library screening . Northern hybridization analysis showed that the cold-induced conditions , cotton 36 CBF gene expression after 4 hours , 4 to 12 h between the expression levels continue to improve . But less than its transcripts were detected in the 24 h . 35S Lee NOS promoter were constructed cotton CBF plant expression vector , and cotton genetic transformation by pollen tube pathway , functional identification and molecular signed in progress .
