Abstract


Objective: generation and accumulation of oxidative DNA damage in various gastric mucosal lesions such significance in the occurrence and development of gastric cancer . Helicobacter pylori (Hp) infection status may lead to a significant increase in gastric mucosal cells endogenous and exogenous oxidative DNA damage related research is currently limited to the use of immunohistochemical method study for various types of Hp-related gastric mucosal lesions and pathological data Helicobacter pylori is difficult to directly reflect on the role of oxidative DNA damage in gastric epithelial cells . In this study, Hp to culture filtrate direct effect on the normal human gastric epithelial cell line GES - 1 , using immunocytochemistry assay treatment gastric epithelial cell GES- 1 within the oxidative DNA damage . Aims to explore the role of oxidative DNA damage induced by Hp Hp-related diseases . Methods: liquid culture Hp strains NCTC11637 and G50 , until at 600nm optical density value reached 0.7 , collected by centrifugation Hp culture filtrate , -80 ℃ save . The vacuolating cytotoxin test is used to detect the presence of Hp culture filtrate vacuolating cytotoxin (VacA) . Tetrazolium salt (MTT) colorimetric test for the detection of Hp culture filtrate of GES-1 cell proliferation . Climbing film culture filtrate of different concentrations of Hp and Hp blank medium role in normal gastric epithelial cell line GES-1, and cells were removed at different times , 4% paraformaldehyde immunocytochemical method for the detection of GES- 1 intracellular oxidative DNA damage sensitive indicator of 8 - OHdG expression . Results : 1 . The vacuolating cytotoxin trial 4,6,8,10 mg / ml the NCTC11637 culture filtrate with Hela cells at 48h , visible under the microscope more than 50% of the Hela cytoplasmic vacuolar degeneration and various concentrations of the G50 culture filtrate acts on Hela cells not
