Abstract


Cerebrovascular disease is one of the major diseases of serious harm to human health . In China, more people died of cerebrovascular disease than heart disease and cancer , the largest in the three major causes of death , including ischemic cerebrovascular disease accounted for 60% to 80 % . Cerebral ischemia caused by a variety of reasons will eventually lead to nerve cell necrosis , patients sequelae troubled . According to the cerebral ischemia caused by nerve cell damage and death of the pathological mechanism of research and development of drugs to inhibit nerve cell necrosis is called a neuron protective agent , including ion channel modifiers, receptor modulating agents , free radical scavengers , epoxidized enzyme inhibitors and the like. NS-7, 4 - (4 - fluorophenyl ) -2 - methyl-6 - (5 - piperidin- pentyloxy) pyrimidine hydrochloride Nippon Shinyaku , Japan developed a novel neuron protection agent. The study found , NS-7 easy to penetrate into the brain , are mainly distributed in the substantive part , it can block the voltage -sensitive sodium channel neurotoxin receptor sites 2 , inhibition of calpain and the of Na < / sup > channel and high - voltage activity L and N-type Ca 2 channel has blocking effects . The protective effect of NS-7 on the injury of rats with hypoxic brain cortex slices confirmed its the of Na < / sup > / Ca 2 channel blocking effects . Closed to occur in the middle cerebral artery in rats injected sufficient amount of NS - 7 , a protective effect on brain ischemia . Local infusion of NS-7 (10μM) will not affect its own dopamine release , but can significantly inhibit the release of dopamine caused by KCl and veratridine given . In addition , NS-7 of Na < / sup> channel and L , P / Q-type Ca 2 < / sup > channel Ca 2 < / sup > flow blocking it produce inhibition of NOS activity .
