Abstract


Mastitis in dairy cows is a frequently-occurring disease, is caused by the loss of the most serious diseases of the dairy industry. Major histocompatibility gene complex (MHC, Major Histocompatibility Complex) is a genetic region on chromosome consisting of highly polymorphic loci by closely chain, their coding MHC antigen in the immune system of the animal plays an extremely important role, and has a very close relationship between livestock disease resistance and susceptibility. Bovine MHC, also known as bovine leukocyte antigen (Bovine Lymphocyte Antigen, BoLA) gene complex on chromosome 23 in cattle, which of BoLA-DRB 3 so far think polymorphism The most abundant gene locus, and is highly expressed antigen molecules responsible for coding class Ⅱ β chain. Subject to BoLA-DRB 3 gene polymorphism research aims to explore the gene as a candidate gene-assisted selection for resistance to mastitis traits feasibility. BoLA-DRB 3 gene polymorphism of this study was analyzed by PCR-RFLP method, the results show that: the gene locus were treated with two restriction enzymes RSA I and Hae Ⅲ digestion, showing highly polymorphic, respectively detected seven alleles and 11 genotypes and two alleles and three genotypes. Rsa Ⅰ AA, AD and Hae Ⅲ AB, AA predominant genotype Rsa Ⅰ A, D and Hae Ⅲ A dominant gene. Multi-factor analysis of variance analysis the Year Quarter BoLA-DRB 3 different genotypes of various milk production traits, the results showed that: Year Quarter BoLA-DRB 3 Rsa Ⅰ different genotypes digested protein percentage and milk fat reached a significant level (P lt; 0.05). The results show that: in terms of milk fat traits Rsa Ⅰ AB, AC, AE, AD, DD, the EH type of individual are significantly higher than the of Rsa Ⅰ EG type (P lt; 0.05 by multiple comparison analysis of different genotypes of all milk production traits ): Rsa Ⅰ AC, AE, DD-individuals are significantly higher than the of Rsa Ⅰ DE type (P lt; 0.05); Rsa I AE-type individuals was significantly higher than the Rsa Ⅰ AA, AD-type (P lt; 0.05). Rsa Ⅰ EG type of individual protein percentage traits, significantly lower than the other Rsa Ⅰ type (P lt; 0.05); Rsa Ⅰ AA, DD, GG genotype individuals significantly with higher than Rsa Ⅰ AB, the AE-type (P lt; 0.05); Rsa Ⅰ DD type individuals were significantly higher than Rsa Ⅰ AD type (P lt; 0.05); individual with Hae Ⅲ in AB significantly higher than the Hae Ⅲ AA type (P lt; 0.05). Of Rsa Ⅰ AD type SCS was significantly higher than the Rsa Ⅰ EG type (P lt; 0.05), Rsa Ⅰ EG type with low SCS significantly associated marker-assisted selection for resistance to mastitis traits.
