Objective: To study the polymethyl methacrylate bone cement (polymethylmethacrylate, PMMA) viscous as early as the mid-viscous late anti-tumor effect. Materials and methods: 1. The paravertebral tumor model: CT-guided percutaneous VX2 tumor inoculated into the paravertebral soft tissue of New Zealand white rabbits, paravertebral tumor models. Inoculated with a half underwent PET-CT examination model, and the determination modeling success paravertebral tumor SUV maximum value (SUVmax) and the SUV average (SUVmean,). 2 experimental groups and treatments: Select 30 successfully establish the paravertebral tumor model the experimental rabbits were randomly divided into A, B, C group, n = 10. Group A the viscous state group for analog PVP bone cement injection, 1.0mlPMMA bone cement is injected into the tumor at the sticky early and mid; B group for analog PKP bone cement injection viscous state group 1.0mlPMMA bone cement late for viscous injection into the tumor; C group sham-operated group, 1.0ml saline injected into the tumor. The first postoperative day, seven days each select five experimental animals, preoperative examination method again to accept the PET-CT examination the Determination of tumor SUVmax, SUVmean value. And experimental animals were sacrificed, take paravertebral HE staining of tumor specimens by TUNEL detection of apoptosis. OUTCOME MEASURES: ① SUV value change: Determination of the experimental group before and after treatment, the paravertebral tumor at each time point the SUVmax, SUVmean value; 2 HE staining results: light microscope intratumoral injection of bone cement, bone cement surrounding well away from bone cement at the tumor cell necrosis; ③ TUNEL apoptosis detection results: observation of tumor cell apoptosis and calculated the apoptotic index. 4. Statistical analysis: using reusable test of variance analysis and comparison of before and after treatment the tumor SUVmax, SUVmean value difference; using factorial analysis comparing treatment the tumors of each group at different time points between SUVmax of SUVmean value with or without differences; analysis using analysis comparing treatment between different groups, different time points, bone cement surrounding and apoptotic index of tumor cells away from the bone cement at whether the differences. Results: 1.PET-CT detection: tumor SUV values ??of group A and group B before treatment and after treatment at each time point were there is a statistically significant difference (P lt; 0.05), the SUV values ??significantly decreased after treatment, to treat reduce the most obvious, the saline group were tumor SUV values ??gradually increased; after treatment, there are statistical differences between each time point tumor SUV values ??of group A and group B and group C (P lt; 0.05); treatment was no significant difference between time points tumor SUV values ??between group A and group B (P gt; 0.05). 2.HE staining showed that: one day after treatment, group A, group B bone the cement surrounding area tumor cells significantly necrosis visible little residual tumor cells in the necrotic tumor cells, especially in the area away from the tumor of the bone cement than more common, and 7 days after treatment, bone cement surrounding residual tumor cells increased significantly compared to the first day, away from the bone cement at the growth of tumor cells active. Saline group at each time point of tumor necrosis obvious. 3.TUNEL apoptosis detection results show: the apoptotic index between different materials group, between the different parts and different time points comparisons with a statistically significant difference (P lt; 0.05). One day after the group A, group B bone cement surrounding the tumor cell apoptosis, tumor cells away from the bone cement visible part of the apoptosis, but the differences between the two groups was not significant. The first 7 days after treatment, tumor cell apoptosis was significantly reduced. The tumors in the control group to see a few small pieces of apoptosis. Conclusion: PMMA bone cement can lead to obvious necrosis of tumor cells in the short term, and can significantly induce tumor cell apoptosis, and its strong anti-tumor effect of neighboring tumor cells in the tumor cells away from the bone cement at. With the anti-tumor effect of time after treatment extend such decline. PMMA bone cement short-term anti-tumor effect in the viscous as early as the mid-and viscous late no difference.
