Local Administration of Interleukin-1 Receptor Antagonist Intervening Achilles Tendon Contracture in Rats: A Empirical Study

Abstract


Objective To observe the interleukin-1 receptor antagonist (IL-1ra) on rat Achilles tendon morphology after complete stress shielding and Ⅰ type III collagen, TGF-β, MMP-1/TIMP-1 expression in, and to explore the interleukin - 1 (IL-1) in the tendon contracture and IL-1ra tendon contracture rats and possible mechanisms. 8 to 10 weeks of age, weight 210 ± 10g, male SD rats were 40, were randomly divided into 2 and 4 weeks Achilles tendon contracture of the model, two weeks and four weeks of IL-1ra intervention group, n = 10, 10 rat right hindlimb composed of normal control group were randomly selected from two weeks making module and the intervention group. Left hindlimb of 40 rats were used Achilles tendon cerclage combined sciatic nerve transection model, postoperative penicillin to prevent infection three days, one week rat box covered with sawdust free stocking a normal diet, and then transferred to the cage within the free-range. Two weeks of the model and intervention groups, respectively, after the operation, after 1 week, respectively after four weeks of the model and the intervention group, after one week, two weeks, three weeks around the Achilles tendon of the left hind local injection of phosphate buffer salt solution (PBS) and the recombinant human IL-1ra (rhIL-1ra). Achilles tendon of the normal control group received no treatment. To 4 weeks after surgery were sacrificed Achilles tendon. Taken at random from each group 2 Achilles fixed with 2% glutaraldehyde solution, the TEM observation, to observe the changes in morphological aspects. Remaining Achilles tendon tissue with liquid nitrogen, grinding the ELISA test. Results 1. Morphology: intervention group (1) naked eye under Achilles tendon than made module small, smooth, but thick compared to the normal group. (2) electric microscope two weeks of the model and the intervention group collagen fibril cross-section with the normal group, similar to, but longitudinal section arranged relaxation, disorder; 4 weeks made modules crosscut surface and longitudinal section surface were with normal significantly with differences, the performance of tiny fibers significantly increase derangement. The intervention group showed a small increase in the fiber cross-section of the normal close, but the longitudinal section seems a little relaxation. Biochemical detection: two weeks of type I collagen levels in rat Achilles tendon stress shielding significantly elevated was no significant difference between the two groups, began to fall to 4 weeks, the model below normal, the intervention group tended to be normal, The difference between the two groups is statistically significant. Type III collagen stress shielding 2 and 4 weeks after the two groups were significantly higher than normal, no significant differences between the two groups, two groups of four weeks compared with 2 weeks significantly decreased. TGF-β content of 2,4 weeks in both groups significantly increased between the two groups had no significant differences, no significant differences between two weeks and four weeks of each group. The MMP-1/TIMP-1 stress shielding after 2 weeks were significantly higher model group was significantly higher than in the intervention group, between the two groups there are significant differences between the model and the intervention group group 4 weeks there was no significant difference. Two 4 weeks, significantly less than two weeks near normal. Conclusion 1. IL-1 plays a key role in the tendon contracture, IL-1ra the effective intervention tendon contracture. Contracture tendon fine fibers increased with IL-1 are closely related. Tendon contracture 3.IL-1ra intervention mechanisms may reduce type I collagen content and increased MMP / TIMP.
