Abstract


Objective: coronary intervention embolism established myocardial infarction animal models , under the guidance of the digital subtraction angiography after coronary injection autologous bone marrow mesenchymal stem cells (MSCs), studies of left ventricular function after myocardial infarction and between fibrosis , and to explore the feasibility of autologous bone marrow mesenchymal stem cell transplantation in the treatment of myocardial infarction . : 10 Taihu Meishan pigs by coronary embolization randomly divided into 2 groups : group A transplant group , infarct 4 weeks after injection of MSCs ; B group , the control group , infarct 4 weeks after injection DMEM ; ( n = 5 ) . The extracted experimental pig iliac bone marrow in vitro isolation , culture , purified mesenchymal stem cells in the bone marrow , number of about 1.0 × 10 ~~ 7 MSCs injected slowly infarction by interventional 4 weeks experimental porcine coronary embolism remote . 1 week, 4 weeks after transplantation observed left ventricular systolic and diastolic function ; detect Four weeks after the infarct zone and non-infarcted myocardium collagen deposition amount ; the conventional HE stained producer , Masson trichrome staining observed infarct zone and border zone the integration of the basic structure of the arrangement and myocardial collagen ; extracted at different points in the experimental animal venous blood after centrifugation serum TGF-β1 and MMP-1 levels . Results: mesenchymal stem cells in the bone marrow 4 weeks after transplantation , LVSP and dp / dtmax significantly improve , -dp/dtmax than pre-transplant , LVEDP significantly lower than the pre-transplant , and compared to the control group, a statistically significant difference (P lt; 0.05) ; transplant groups infarction Center area and non - infarct zone collagen deposition amount was significantly lower than the control group , in which the noninfarcted collagen deposition than transplants decreased , a statistically significant difference ( P lt; 0.05 ) . Stem cell transplantation at different time points after serum TGF-β1 content declining but amounted to 2 weeks when the valley is no longer reduced (P lt ; 0.05 ) , the control group, only a slight downward trend; Serum MMP- one week after the stem cell transplantation 1 content increased, reached a peak after 2 weeks ( P lt; 0.05 ) , while the control group remained stable at
