Abstract


Objective To investigate the Salmonella typhi Type IVB pili whether to participate in the inflammatory damage caused by the bacteria in the body , whether to activate PKC, ERK, NF-κB signaling pathway induced THP-1 cells the expression of IL-6 . THP-1 cells with Type IVB pili A21 - 6 bacteria , missing IVB pili the Pils bacteria were incubated ; ELISA assay of IL-6 production by RT - PCR detection of IL-6 mRNA expression levels of , non- radioactivity assay detecting changes in PKC activity detected by Western blot ERK phosphorylation levels of luciferase reporter gene analysis detection of NF -kappa B activation of expression . Results of THP-1 cells were treated with A21 - 6 the IVB type pili bacteria stimulate IL - 6 and the generated level , mRNA expression levels of PKC activity changes , ERK kinase phosphorylation level and the expression of NF - kappa B activation was significantly high in the pils bacteria group ; PKC , ERK and NF - kappaB activation levels were stimulate 10min , 30min and 60min after significantly increased ; and in 30 minutes , 60min and 120min about peak . IVB pili can be suppressed to a certain extent on the PKC inhibitor DECA -induced THP-1 cells PKC activity , NF - kappaB activation expression of the IL-6 level . Conclusion Salmonella typhi Type IVB pili significantly enhanced THP-1 cells PKC , ERK and NF - kappaB activation levels the IVB of pili in Salmonella typhi induced monocyte / macrophage cell PKC , ERK and NF - kappaB activation process is a kinds of strong stimulus ; IVB pili of Salmonella typhi induced monocytes / macrophages to produce IL-6 process is also an important stimulus ; PKC , ERK and NF - kappaB three kinds of signaling pathways involved in the molecular process of intracellular signal transduction . Salmonella typhi Type IVB pili may be caused by activation of PKC → ERK → NF - kappaB pathway synthesis and release of IL-6 and other proinflammatory cytokines , resulting in pathological damage of the body .
