Abstract


Soy isoflavones is the soybean growth process of the formation of secondary metabolites , a mixture of polyphenols , with a variety of biological activity , especially anti-cancer research has become the world's Top focus . This paper studies the isoflavones extracted from soy germ meal , and further separation of refined higher technology content products . First , the establishment of the method of HPLC analysis of the isoflavones . Analysis conditions : Novapak C 18 column (φ3.9 × 150mm, 4μm), mobile phase: A - 0.1% acetic acid / water , B - 0.1% acetic acid / acetonitrile , gradient elution ; flow rate of 1.0mL/min, column temperature was 40 ° C , and the detection wavelength was 254 nm . Isoflavone extraction, diafiltration , 60% ethanol, the flow rate 1.0BV / h , 50 ° C The operating conditions , the the germ bulk density of 0.26g/mL, ethanol used in an amount of 4.0 ml / g germ obtained under this condition crude extracts of total isoflavone content of 6.43 % . Column chromatography purified isoflavone crude , but before this , in order to reduce the malonyl isoflavone loss of the process in the column chromatography , the first the isoflavones stability under high temperature conditions to test , and to determine the high temperature hydrolysis conditions for 100 ℃, hydrolysis 5h. Malonyl complete hydrolysis of acetyl , unconjugated aglycone isoflavone . Polyamide resin chromatography media , dynamic adsorption and desorption process . Affect the adsorption effect of flow rate , temperature and concentration of the sample were investigated adsorption process : the chromatography column specifications φ1.2 × 53cm, 40 ℃, on sample concentration 5mg/mL , flow rate 1BV / h under the conditions of saturated adsorption amount , polyamide isoflavone of 60.0mg / g, penetration adsorption amount of 25.0mg / g, the mass transfer zone at a height of 30.0cm ; desorption of the isoflavone to the content of isoflavone products and isoflavone recovery indicators, inspect the desorption solvent concentration, temperature and amount of the solvent , and other factors to determine the desorption operation conditions were as follows : 40 ℃, velocity 1BV / h , the eluent is water 1BV , 50 % ethanol 1BV , 95 % the ethanol 2BV, collected as 2.25 ~ 2.92BV, product content of 41.11% , isoflavones recovery rate of 76.46% .
