The Observation on the Ultrastructure of Bioartificial Aponeurosis under Electronic Microscope

Objective To observe the ultrastructure of bioartificial aponeurosis under electronic microscope in the assimilation process in rats, and to investigate the changes in the process.Methods The animal aponeurosis defect models were established. The bioartificial aponeurosis, the autologous aponeurosis, and the homologous aponeurosis were transplanted into the defects, respectively, and were taken out at the weeks 2, 4, 8, and 12 after transplantation. The histological changes of the transplanted aponeurosis were observed with electronic microscopes, and the results were compared among the 3 groups.Results The bioartificial aponeurosis showed no cell components and dense fibers under electric microscope at the 2 weeks after the transplantation.Macrophages were observed in peripheral tissues, with many lysosomes in the cytoplasm. At the 4 weeks after the transplantation,no fibroblasts were observed, but the fibers of the bioartificial aponeurosis were breaken, with dense macrophages and fibroblasts attaching to its breaks. At the 8 weeks after the transplantation, The bioartificial aponeurosis were absorbed, where there were abundant rough endoplasmic reticulum, fibroblasts, and collagen fibrils of different sizes. Macrophages decreased. At the 12 weeks after the transplantation, the fibroblasts were flat. The nucleus were small, and oval in shape. The rough endoplasmic reticulum, free ribosomes and Golgi’s complex were declining in the cytoplasm, which showed the declining functions of proteins. The histological structure, fibroblast structure and macrophage structure of the 3 kinds of aponeuroses were of no statistical difference under electronic microscope.Conclusion The bioartificial aponeurosis is not irritant to the organisms. Its histological structure gradually turns into the structure of auto-aponeurosis. The material has good tissue assimilation, and can be used as a satisfactory replacement in the treatment of aponeurosis defects.
