The Significance of Expression of Endothelin-1 and Its Receptor in Renal Cell Carcinoma

Abstract


Objective To detect the expression of ET-1 and its receptor in renal neoplasm and investigate the relationship with cell apoptosis and cell proliferation, explore the possible mechanism of ET-1 influencing renal cell carcinoma.Methods Immunohistochemistry SABC was applied to detect ET-1 and its receptor expression in 10 normal kidney samples and 46 patients with renal cell carcinoma, the same way was used to check up PCNA, and renal cell apoptosis was evaluated by means of terminal deoxynucleotidyl-transferase (TDT) mediated dUTP-biotin nick end labeling technique (TUNED .Results The protein expression of ET-1 and ETAR in renal carcinoma was mostly located in the renal cell plasma, the positive rate was 45.65% and 80.43%, respectively. It was more then that in normal kidney samples (30% ) . The level of ETAR was correlation with the grade and the prognosis of renal cell carcinoma (P<0.05), but there was no significant difference between the clinical stages. The expression of ET-1 was negatively correlated with cell apoptosis and cell proliferation.Conclusions The association of ETAR expression with cell apoptosis and cell proliferation indicated ET-1 protein inducing carcinoma by suppressing cell apoptosis and increasing cell proliferation in autocrine and/or paracrine way. The assessment of theexpression of ETAR protein would be one of the important indexes of the development and prognosis of renal cell carcinoma in the future.
