The Preliminary Research of the Effect of External Urokinase-type Plasminogen Activator(uPA) to the Synovial Tissue of Rabbits’ Keen Joint

Abstract


Methods The 42 New Zealand white rabbits were devided into 2 groups as random, 24 experimental, 18 matched control. The experimental animals were injected intraarticularly with uPA in right knee joint, control animals were injected with the same dose fo normal saline. All animals were killed in 4,8,12 weeks respectively after the injections. The weight of the rabbits were messured,the synovial tissue of the both side of the joints were taken to carry on histology observation(HE) and immuno- histochemical check.Results The knee joint’s synovium of both sides of the the control group were velvet ,no tumefaction and incrassation. The knee joint’s synovium of both sides of the experimental group of 4 weekes were found swelling adhension; overgrowth,conglutinated obviously of 8 weeks; fibrosis of tissue below the synovium was obviously of 12 weeks by eyes. From the HE coloration by microscope, the knee joint’s synovium of both sides of the experimental group of 4 weekes were at the condition of acut inflammation, neutal cells were the mostly cell, the synovial cells were overgrowthed like papilla obviously, fibrosis of tissue below the synovium was not serious. Of the 8 weeks, acut and cronic inflammation exsited of synovium;the neutral cells and lymphocyte were all found;the blood vessels were overgrothed and hyperaemic significantly; fibrosis of tissue below the synovium was obvious than that of 4 weeks. Of the 12 weeks,the inflammation of synovium was chronic and rehabilited change,the lymphocyte was the primary inflammation cells,the lymphocyte follicle was appeared;the hyperplasia of blood vessels was still obvious;the fibrosis of tissue below the synovium was the most obvious of the three groups.These changes were significant difference between the experimental group and the control.From the immunohistology ,the difference was highly significant(P<0.05) of both sides of joints of every period of the experimental and the control group.There was no difference (P>0.05) between the injected an not injected joint of the experimental group. There was no difference (P>0.05) between the injected an not injected joint of the contol group. The analysis of MVD show that there was highly significant(P<0.05) difference of both sides of joints of every period of the experimental and the control group. There was no difference (P>0.05) between the injected an not injected joint of the contol group. The analysis of weight show that here was highly significant(P<0.05) difference of every period of between the experimental and the control group.
