Abstract


On the basis of predecessor research,the study is on the basis of supplementation of glucose 3-3(glucose-citrate- yelk),again supplementation of fish oil enriched PUFA and VE in frozen sheep semen as the new additive, furthermore by testing a series of indexes, researching the protected effect of PUFA to the sheep frozen- thawed semen and the effect of PUFA to sheep frozen- thawed semen fecundation rate.The objective of the research is the effect on sheep frozen ram semen diluent with supplementation of different proportion VE and PUFA ,after frozen-thawed,test usual indexes such as thawed semen motility and intact acrosome ,and Ca2+、Na+、Cl-、LDH enzyme vitality and GOT enzyme vitality etc; at the same time after comparison, ascertain the optimum mixture ration and test thawed semen and fresh semen ultrastructure by transmission electron microscope.The results show that:1. The effect of supplementation of 1% VE and 3% PUFA in sheep thawed semen diluent is less to the sperms harm than the control one after frozen-thawed.every index is all improved after thaw, semen motility and intact acrosome rate are increased respectively by 6% and 10% than the control one(P<0.05),LDH enzyme vitality and GOT enzyme vitality of the semen are reduced respectively by 65% and 15% (P<0.05);2.The semen motility and intact acrosome rate are significantly increased than the control one(P<0.05) with supplementation of PUFA 0.5%、1%、2%、3%、5% after thaw;at the same time,there is a increasing tendency of the semen motility and intact acrosome rate after thaw while PUFA concentration is increased;but which are comparatively lower than the treatment group of supplementation VE ;3. The semen motility(0.345) and intact acrosome rate(0.396)are significantly increased than the other treatment groups with the supplementation of 1% VE in sheep frozen semen diluent and the supplementation of 3% PUFA in the diluent by 2-step method of cryopreservation procedure(P<0.05), LDH enzyme vitality and GOT enzyme vitality of the semen are significantly reduced than the other treatment groups (P<0.05);4. The semen motility(0.328) and intact acrosome rate(0.335)are significantly increased than the control group and the other treatment ones with the supplementation of 3% VE in sheep frozen semen diluent and the supplementation of 3% PUFA in the diluent by 2-step method of cryopreservation procedure(P<0.05), LDH enzyme vitality and GOT
