Enzyme immobilization is one of the most important research fields in enzyme engineering and is cared more and more. Lipase(EC, acylglycerol hydrolase) which is widely used in the hydrolyzation, alcoholysis and the synthesis of ester, lactone, polypeptides, macromolecular polymer and the splitting reaction of enantiomorph polymer in the interface of oil-water. However, most free lipases are insoluble and easy to get together in organic solvents. And free lipase itself is not stable and hard to be separated from the reacting system. It helps to widen the interface between enzyme and reactant ,diffuse the reactant, enhance the thermal stability of enzyme, regulate and control the activity and stability to immobilize the enzyme. And the immobilized enzyme also benefits the reaction in the organic solvents and its separation from the products. Besides, it can be reused several times. These strongpoints of immobilized enzyme also play important roles for improving its catalyzing efficiency and prolonging the time of being used.Mesoporous Molecular Sieve MCM-41is a kind of inorganic porous material which has many priorities such as outstanding stability, high mechanical intensity, being nontoxic to microorganism, to resist the acid and the alkali, low cost, long life-span and so on. Because of these good characteristics, MCM-41 is taking on promising future in enzyme immobilization.According to the property and the use of enzyme, the methods for immobilizing enzyme mainly include adsorption, crosslinking, covalent binding and entrapping. The synthesis of mesoporous molecular sieve MCM-41 and immobilizing the lipase on it by simple physical adsorption was studied in the paper. The influence on the enzyme activity and recovery under different immobilizing conditions was also studied. Besides, the thermal stability and the operational stability of immobilized enzyme were discussed.
