Abstract

In the modern society, many people have to do long time continuous work, for example the long-distance driver drives, the continual military operationsly and so on. It is well known that these ultra dilirnal periods work as well as compulsory work without enough sleep can inevitably have the serious influence to operator’s capacity and the cognitive function, which usually reduces the working efficiency. In the present study, we have successfully established a rat model of long-term continuous operations, which can reasonably simulate combined influences in the long-term continuous operations circumstance. Subsequently, we studied the deleterious effects of long-term continuous operations on rat awake activity. Then we investigate the effects of long term continuous operations on orexin-A and its receptor (OX1R) expression. After the long term continuous rat model is injected with coffeine we investigate the expression of orexin-A and its receptor (OX1R) .1. Establishment of a rat model of long-term continuous operationsRat model was established by using a treadmill protocol. Rats were allowed to do continous exercise for 48 h in the conventional treadmill equipment used for small animal. We suggested that the treadmill protocol was an available way for simulating combined influences in long-term continuous operations circumstance, such as long-term physical exercise, intellectual activity caused by avoiding the electricity stimuli (conditional reflex), stress factors, food and water limitation and sleep deprivation and so on. Thus our model has successfully simulated many kinds of complicated environment in long-term continous operations.2. Effects of long-term continous operations on rat awake activity during the nightIn this study, rat awake activity during the night was evaluated by three indexes, including distribution of behavioural activities, wakefulness time and rearing number. Our data showed that long-term continous operations markedly decreased the distribution of behavioral activities (n=8,P<0.05), wakefulness time (n=8,P<0.01) and rearing number (n=8,P<0.01). Thus these results have illuminated that long-term continuous operations have a deleterious effect on rat
