Abstract


With the changes of life style, nonalcoholic fatty liver disease (NAFLD) has been on the increase and turns out to be an important disease in many developed and developing countries. In China, its incidence is gradually increasing too. So far, the pathogenesis of NAFLD has not been identified. Yet, previous studies have shown that fatty acid metabolic disorders are foundation and key link in the development of NAFLD. It is known that liver is the center in the lipid metabolism that has very complicated regulation. Recently, it has been found that liver X receptorα(LXRα) and sterol regulatory element binding protein-1c (SREBP-1c) are considerable nuclear factors in the regulation of lipid metabolism, and the key to the regulation of fatty acid biosynthsis. Thus, the investigation of the role of LXRαand SREBP-1c in the development of NAFLD can conduced to a coprehensive knowledge of the pathogenesis of NAFLD.In this study, the model was established, in which rats’NAFLD was induced by fatty diet. Through this model, the general shape and histopathology of liver was observed; the concentration of free fatty acid (FFA), triglyceride (TG), alamine aminotrasferase (ALT), aspartate aminotrasferase (AST) in serum was detected; the expression of SREBP-1c in liver tissue were measured with immunohistochemistry (IHC); the mRNA and protein level of LXRαand SREBP-1c were detected by reverse transcription polymerase chain reaction (RT-PCR) and Western blot; and the activity of fatty acid synthase (FAS) was measured by ultraviolet spectrophotometer. The aim of this study was to explor the possible mechanism of LXRα, SREBP-1c in the development of NAFLD from the perspective of fatty acid metabolism. The main results and conclusions are as follows:1. After establishing the model in which the rats’NAFLD was fed by fatty diet, the general shape and histopathology of liver was observed, and the concentration of FFA, TG, ALT and AST in serum was detected.
