The shoot-tips of Morus wittiorum Handel-Mazzetti were used as explants in tissue culture for rapid propagation. And then the method of colchicines inducement combining tissue culture technology has been used to gaining the new polyploidy germplasm of Morus wittiorum Handel-Mazzetti. Additionally, tetraploid germplasm has been obtained which can be used in the breeding of mulberry.The results of experiment as follows:The results in the tissue culture of the Morus wittiorum Handel-Mazzetti showed that: the optimum sterilization method was 10 minutes immersing by scour, 30 minutes washing by water, 40 seconds sterilizing by 75% ethanol, 12 minutes sterilizing by 0.1% mercuric chloride, and washing 4-6 times by sterilized water. The inducing rate of bud initiation is 90%, and the contaminated rate is only 10%, the time of sprouting is 20~27 days. The results indicats that MS medium supplemented with BA 2.0 mg/L + NAA 0.2mg/L + sucrose 30 g/L is suitable for multiplication and 1/2MS+BA 0.1 mg/L+NAA 0.05 mg/L+sucrose 30 g/L is optimal for rooting. And the best rooting rate is 97.4%, the average of each plant has 6.2 roots.On the basis of building the system of in vitro regeneration, the inducing tetraploids of the Morus wittiorum Handel-Mazzetti was carried out by the solution of colchicines. Three methods were adapted: immersing the buds in different concentration solution of colchicines, inoculating buds in the solid medium supplemented with colchicines and in the liquid medium supplemented with colchicines. The results showed that: the method of immersing buds was better than the others, the highest inducing rate was 26.7%. The autopolyploid could be most effectively induced by immersing buds in 0.2% colchicines for 3 days. The optimum concentration of inducing for all the methods was 0.2%.Plant morphology observation of Morus wittiorum Handel-Mazzetti with similar growth stage and environment background suggested that tetraploids are different from corresponding diploids in the forms of leaves, In general, the length and width and thickness of the leaves of tetraploids are bigger than those of diploids, but with smaller leaf index. The characteristic of tetraploids stoma with that of diploid was compared. Stomata of polyploids are larger and spaced father apart on the undersurface of the diploids with less stoma density which is 60.9% than that of diploids.
