Studies on Toxic Effects of F-2 Toxin and Protective Effects of Vitamin E on Cultured Ovary Granulosa Cells

F-2 toxin is a raycotoxin, commonly produced by various Fusarium species. F-2 toxin has strong estrogenic activity inducing hyperestrogenism and physiological alteration of the reproductive tract. Moreover, it was shown to be genetoxic and immunotoxic. F-2 toxin can decrease the quality of animal and poultry products, and can also cause serious health problems in humans. So researches are necessary to be carried out on toxin effects of F-2 toxin and its detoxification.Research Subject: It is to discuss the toxicity and function mechanism of F-2 toxin on the ovary granulose cells, and vitamin E is used as an antipoison to clarify the detoxification mechanism of F-2 toxin, which will provide theoretic basis on manufacturing an effective antipoison which can wipe off the mycotoxins found in feed and foodstuff.Research Methods: Swine ovary granulosa cells are used as empirical model. Swine ovary granulose cells were isolated by disposable syringe. The toxin groups were treated with F-2 toxin at different concentrations (80 μg/ml, 20 μg/ml, 5 μg/ml, 1. 25 μg/ml), the detoxication groups were added vitamin E(50 mg/ml) and F-2 toxin at different concentrations (80 μg/ml, 20 μg/ml, 5 μg/ml, 1. 25 μg/ml), the control groups were treated with cultured fluid. Injury and protection of cells was detected by MTT assay. The DNA fragmentation was detected by the single cell gel electrophoresis. The activity of SOD in ovary granulose cell and in the cultured solution was detected by xanthine oxidase assay. The value of MDA in cell cultured solution was detected by TBA assay. The apoptosis of ovary granulose cell was detedcted by DNA agarose gel electrophoresis.Research Result: survival percent of isolated ovary granulose cells was from 85% to 95%. The proliferation of the cells was inhibited by 5 Mg/ml、 20 μg/ml, 80 μg/ml F-2 toxin in dose-dependent and time-dependent. 20 Mg/ml、 80 μg/ml F-2 toxin resulted DNA fragmentation, and the tail rate was 82.67% and 80.33% separately.
