Abstract


Baoxinkang has good effect in treating congestive heart failure patients of deficiency of both Qi and Yang.To study Baoxingkang’s pharmacological role,the present research was preformed,in which the sequent changes of Bax protein and Bcl-2 protein expression of left ventricular myocytes were observed in pressure overloaded rats.And the role of Baoxinkang on the two proteins was also studied.The present study included two parts:the first part was the observation of expression of bax and bcl-2 gene,regulating apoptosis,in pressure overloaded rats.produced by obdominal aortic constriction(AC),Bax and Bcl-2 were detected at 1,4,7,10,14,21,30 and 40 days after operation.In the second part of the experient,the rats was decided into three groups:Baoxinkang group(BXK),distilled water group(DW) and sham-operation group(SH),the rats were killed at the day when the ratio of Bax and Bcl-2 protein was the highest according to the first part experient.and Baoxinkang’s effect was studied.The expression of Bax and Bcl-2 protein was detected by flow cytometry.In the experient,the myocardial tissue was isolated single cardiomyocytes successly, in addition, the method of detecting the intramyocytal antigen was created.Results:In the first part.l day after AS.the expression ratio of Bax protein in left ventricular(LV) myocytes was higher(63.75%),that of Bcl-2 protein was lower(31.45%),the ratio of Bax and Bcl-2 protein(Bax/Bcl-2) was 2.02,the expression ratio of Bax protein didn’t vary with the time.it was between 50% to 60%;but the expression ratio of Bcl-2 protein was different to that of Bax,it increased to the highest at the 7th day,sequently,it declined slowly,and at the 30th day,it dropped to the lowest,22.8%;Bax/Bcl-2 decreased from the 4th day to the 7th day.it increased sequently.and reached to the highest at the 30th day,Bax/Bcl-2 at the 40th day was lower than the 30th day.The 2nd part experient:both the cardiac weight(CW) and left ventricular weight(LVW) of DW rats increased significantly,but that of BXK rats had no significant difference from that of the SH rats,the cardiac mass index(CMI) and the left ventricular mass index(LVMI) had the same result.the expression ratio of Bax of BXK rats was the same as that of SH rats,but the expression of Bcl-2 was lower than that of SH rats significantly(P<0.05),Bax/Bcl-2 was higher than that of SH rats notably(P<0.01);Compared to DW,the two proteins and their ratio of BXK had significant difference(P<0.05 or PO.01), the expression of the two proteins were not different from that of SH(P>0.05),but it had a contradictory result in Bax/Bcl-2(P<0.05).The CW and LVW of female rats were lighter than that of male rats in SH and DW,female rats of SHhad a smaller CWI than male rats,but in DW,the CWI was the same;the expression of Bax of female rats had no difference from that of male rats in SH and DW,but to the expression of Bcl-2 proteinjt was reverse;Although the expression of Bcl-2 protein increased in both female and male BXK rats’ LV cells,and the expression of Bax protein deceased,the expression of Bax in male rats was remarkably lower than that of female rats.Conclusion:!.In the cardiomyocytes of congestive heart failure rats model, Bax/Bcl-2 declined from the 4th day to the 7th day,it rose sequently,and reached the highest at the 30th day,Bax/Bcl-2 at the 40th day was lower than the 30th day,so it was possible that the maximum apoptosis of left ventricular myocells was happened at the 30th day;2.Compared to male rats,it is possible that female rats had better abilities of adaptation and compensation or have lower apoptotic ratio or both of two;3.Baoxinkang could prevent left ventricular hypertrophy which is produced by pressure overload in rats;4.Baoxinkang could upregulate the expression of bcl-2 gene and domnregulate the expression of bax gene in LV myocytes of congestive heart failure rats produced by obdominal aortic constriction,at the same time,it could decrease Bax/Bcl-2,through which it could inhibit cardiac cellular apoptosis, perhaps,this effect is better fo
