Abstract

As a kind of widely distributed plants, the Orchids (Orchidaceae) have high value in ornament and economy. Many people were absorbed in the study on the symbiotic mycorrhizae of Orchidaceae for its vital function to the natural growth of Orchidaceae. There are so many research works having been done on the Orchid mycorrhizae today while few works on mycorrhizae isolated from Chinese Orchids (Cymbidium) which are one kind of most important ornament plants of China. To the native research which with a relatively lag beginning, few emphases were put on the mycorrhizae of Cymbidium and most of the reports are scattered and unsystematic. This paper presented morphological identification study and emphased on the molecular biology research based on AFLP dendrograms of the mycorrhizal strains which isolated from Chinese orchids (Cymbidium). The results are as follows:1. 19 representative mycorrhizal strains were isolated from Chinese orchids (Cymbidium) of 7 species distributed in different sites, all stains were identified as generalized Rhizoctonia fungi according to the colonial morphology characteristics and the hyphae morphological characteristics; 11 different mycorrhizal stains were identified as the anamorphic genera—Euphoria according to the dolipore septum ultrastructure; The results of identification suggested that fungi belong to Euphoria might be the most general species which can develop symbiosis relationship with Chinese orchids (Cymbidium).2. Selection-tests for optimum conditions of mycorrhiza fungi isolation showed that, giving appropriate surface sterilization, taking the mid-part of one-year healthy roots as the isolation materials will obtain the fine isolation results.3. AFLP analysis of 19 mycorrhizal strains based on the dendrograms indicated that, diversity between the symbiotic mycorrhizae of Chinese orchids (Cymbidium) was affected by 3 factors: the environments where orchids distributes, the species and the
