The Screening of Microbial Flora with the Ability to Degrade Cellulose Filter and the Preliminary Study of Its Nature

Abstract


In this study, samples of cellulose degrading microbial flora were obtained from rhizosphere soil, composted wheat straw, and corruption towel gourd. With the technology of regularly altering the regeneration method during on the modified PCS culture medium, 2 groups of mixed flora named No.5 and No.6, with stable and strong capability, were selected by comparing the efficiency of degrading the filter paper. This study also found that the filter degradation efficiency of flora is much higher under shaking culture conditions than static culture. Under 6 days shaking culture in 30℃, 120rpm, 27.96% and 34.21% of the filter paper were degraded by No.5 and No.6.Under microscopy, the two mixed flora were found to be bacteria, and the bacterial characteristics of them are distinct different in morphology: No.5 is mainly found in Corynebacterium, and No.6 in Streptococcus. The change of pH in culture of the two flora were found, with the growth of the culture time, pH increased slowly. After 6 ds, pH in culture of No.5 and No.6 came to stable values around 8.71 and 8.75. And at the same time, the efficient cellulose degradation by the two flora was remined. Research showed wide range of pH in the adaptation of the two flora.Compared different DNA extraction methods, DNA in different periods genomic of No.5 and No.6 flora were extracted. PCR selected the primers F341 and R518 in the V3 area as primers with“GC-clamp”. By comparison of the DNA bands of 16S rDNA with the denaturing gradient gel electrophoresis (DGGE), it’s showed that the structure of the two flora both remained unchanged. No.5、No.6 flora are demonstrated to be efficient and stable microorganisms for the biodegradation of solid waste straw.In this study, flora with the ability of the bacterial degradation of cellulose were obtained directly from the environment, the microbial synergistic effect was effectively full use, avoiding the deficiencies of traditional methods, and providing a good impetus for the recycle of waste straw.
