Abstract


In this paper,Doxazosin Mesylate(DOX) was selected as the model drug to prepare hydrophilic matrix tablets and bi-layer osmotic pump tablets(BOPT).And the drug release character and mechanism were studied. At last,the pharmacokinetic study of DOX hydrophilic matrix tablets was performed.In the formulation study of hydrophilic matrix tab lets, hydroxypropyl methylcelluse(HPMC) and lactose were selected as the main materials to make the tablets.The effects of many factors including formulation and technique on the release behavior of tablets were investigated.Based on the comprehensive single-factor experiments, amount and viscosity of HPMC, sort of adhesives were selected as the causal factors.The formulation optimized by orthogonal experiment design was proved to have the similar release behavior as the imported tablet and its quality was good.The mechanism of the drug release was studied and the result showed that the self-made matrix tablets had the excellent zero-order release character in vitro and their main release mechanisms were matrix erosion.In order to vertify the possibility of DOX made to BOPT ,the formulation and the technique were also studied.In the preparation of
