Abstract


TNFα for fulminant hepatic failure in mice brain AQP4 protein expression Foreword hepatitis easily complicated by hepatic encephalopathy , more than 80 % of patients with cerebral edema, severe hepatitis patients is the direct cause of death in the pathogenesis of cerebral edema not very clear. Some studies show that aquaporin and pathophysiology of cerebral edema associated . In normal brain tissue , AQP-4 is particularly important aquaporins . The purpose of this study was to investigate TNFα in brain tissue against hepatitis AQP-4 expression and its significance. Materials and Methods 1 . Animals and groups : male Balb / c mice , 6-8 weeks old, weighing 18-22g, SPF , were randomly divided into eight groups. Saline control group ; LPS control group ; GalN control group ; LPS liver failure group (LPS GalN) 2,6,9 h; TNFα liver failure group (TNFα GalN) 2,6,9 h; TNFα antibody group ( anti- TNFα-IgG antibody TNFα GalN); anti- TNFα-R1 antibody group : ( anti- TNFα-R1 antibody TNFα GalN); TNFα group . 2 . Model by intraperitoneal injection GalN, TNFα, Lps establish FHF mouse model . Injection of GalN / TNFα via the tail vein 30 minutes before the injection of anti- TNFα-IgG antibody and anti TNFα-R1 antibody. The intended point in time were killed by decapitation and brain tissue in mice livers .
