Abstract

Phenol is an important organic chemical raw materials, downstream products covered many areas the very versatile industrial pollution caused by the environment is extremely serious. Spiny Moina (Moina macrocopa) for the experimental material, based on the observation and analysis of its external morphology and nutrients, phenol stress the Moina intestinal microvilli morphology, digestive enzyme activity, fat metabolism, as well as changes in amino acid composition, aims to explore Moina poisoning digestive function change and adaptation mechanisms, to provide a scientific basis for the toxic mechanism of phenol. Experiments in the scanning electron microscope on the spiny Moina external morphology were observed in detail, using biochemical methods and its nutrients, including general biochemical composition and amino acid composition. The acute toxicity test method blank control group and 5 phenol exposure group (of 0.25,0.75,1.25,1.75 and the 2.25 mg / L). Histology and electron microscopy techniques, treatment of phenol (0.25,1.25 and 2.25 mg / L) for 48h intestinal microscopic and submicroscopic structure was observed. 24h and 48h was determined by spectrophotometry digestive enzyme activity, fat metabolism indicators, including the activities of digestive enzymes including amylase (Amylase), lipase (Lypase) of trypsin (Trypsinase) and protease (Protease and phenol treatment) vitality; fat metabolism indicators, including the content of free fatty acid (FFA), triglyceride (TG) content and lipoprotein lipase (LPL) activity. Liquid chromatography 0.25,1.25 and 2.25 mg / L of phenol for 48h amino acid composition. The results showed that: (1) spiny the Moina female takes the shape of an oval body transparent. 100g spiny Moina wet like moisture, crude protein, crude fat and ash content are 90.92,6.38,1.11 g and 0.67 g. Spiny Moina 17 amino acids accounted for 54.44% of the protein content of the experiment was not detected tryptophan. (2) light microscope, the phenol treatment of spiny Moina the striate reason smooth becomes rough, or even fall off. Electron microscope can be clearly observed, the low concentration of phenol treatment Moina intestinal epithelial cells arranged in a slightly irregular. In the concentration of phenol treatment, microvilli some shedding and breakage, sparse arrangement. Moina by the high concentration of phenol treatment of intestinal microvilli arranged messy, the gap increased, irregular shape. With the increase of the concentration of phenol, Moina intestinal epithelial cells and microvilli worsened the extent of the damage. (3) With the extension of the phenol concentration increased and the processing time, amylase, lipase, trypsin and protease four Digestive enzyme activity showed a different trend, wherein the phenol no significant effect on stress on amylase activity; lipase vigor in phenol 0.25 and 0.75mg / L for 24h compared with the control group significantly (P lt; 0.05) rise; trypsin activity in 24h and 48h are concentration group significant changes, 0.75mg / L concentration group treatment 48h significantly liter high (P lt; 0.05); protease activity in phenol stress, showing first and then decreased with increasing concentration trends, 0.75mg / L concentration group process and 48h reached maximum (P lt; 0.01). (4) With the increase of the concentration of phenol, free fatty acids and triglycerides showed a gradual downward trend (P lt; 0.05); lipoprotein lipase activity in 24h 0.75mg / L concentration group and 48h, 1.25mg / The highest value of L group decreased gradually. Also, for low (0.25mg / L), (1.25 mg / L), high (2.25mg / L) concentrations amino acid composition was measured after 48h in vivo, where there are significant changes in individual amino acids with the control, but the total content and the control there was no significant difference. Conclusion: (1) spiny Moina higher protein content as a common species in Taiyuan, in its fortified can fully meet the growth needs of Yuxia Xie class, excellent creatures live bait. (2) in the intestinal spiny Moina digestion and absorption of the main parts, phenol exposure causes irreversible damage to the intestinal microvilli, thereby affecting the digestive function of Moina adversely affect growth. (3) phenol stress, spiny the Moina assimilation is suppressed; Moina produce more energy through the to strengthen digestion residues in food in the body's ability to maintain normal metabolism. Digestive enzyme activity as Moina poisoning potential biochemical indicators. (4) fat metabolism, is bound to affect of Moina energy metabolism, its growth and reproduction disruption will adversely affect the entire population.
