Abstract


The use of somatic cell cloning technology can bulk copy outstanding boars, rapid breeding of excellent individuals. Abroad has been a useful attempt boars propagation. However, boars cloning efficiency and clone pigs cultivate there is still a big problem. The study of the first cleavage and pig parthenogenetic development (parthenogen-esis, PA) and SCNT (somatic cell nuclear transfer, SCNT) two of embryonic development potential relationship, find the quality of cloned embryos in early developmental important identification of the stage, to further improve the somatic cell cloning technology platform, the cloned pigs efficiency; physical weakness for newborn cloned piglets low survival rate, strengthen management, greater numbers of cloned boars, to establish a The excellent practical boars sets breeding system quickly. The early cleavage PA embryonic development to the the 6d blastocyst rate significantly higher than the mid-late, and the control group (P lt; 0.05; 54.2 ± 3.9% vs.19.7 ± 1.4%, and 5.5 ± 0.6%, 18.9 ± 2.3% ); expansion blastocyst rates also have the same developmental trends. Test: In order to effectively evaluate the the late developmental competence of pig early SCNT embryos, this study to explore the relationship between the first cleavage matter of time before the developmental potential of pig SCNT embryos. The results show that: the early cleavage SCNT embryonic development to 6d blastocyst rate in the mid-cleavage embryos (32.6 ± 3.9% vs.24.6 ± 2.8%), while the lowest rate of late cleavage embryo development to the blastocyst (4.0 ± 0.4%). Early cleavage SCNT embryo development to the the first 6d expansion blastocyst rate significantly higher than the other groups (P lt; 0.05; 19.0 ± 3.9% vs.6.0 ± 4.8% ± 0.2.7.4 ± 0.8, .0.8). Blastocyst cells showed a downward trend between the three groups in the early, middle and late. Test: embryo transfer is an important part of the development status of the embryo late in vivo cloning technology sectors observed. To further illustrate the the late developmental competence of somatic cell nuclear transfer early cleavage embryos, the trials in transplantation link will be early cleavage the SCNT embryo (the first group) transplanted with the unselected embryos in the control group (second group), respectively, observed in vivo development. The results show: the first group of 718 early cleavage embryos were transplanted into 6 receptor sows farrowing 28 (a stillbirth), litter size up to 11; second group of 2719 unselected embryos ported to 12 sows farrowing 25 (8 stillbirths, the three mummies). Receptors in transplantation the early cleavage SCNT embryos were significantly higher than the average litter size and cloning efficiency the unselected embryos receptor (P lt; 0.05; 4.7 vs.2.1; 3.9% vs.0.9%), indicating that early the cleavage embryo implantation after stronger in vivo development. These results show that: the use of somatic cell cloning technology can quickly breeding outstanding boars; initial cleavage as pig cloned embryos growth potential identity effectively predict the late development of the early embryo; the select early bifida SCNT embryos transplant significantly improved cloning efficiency, and provides an important technical reference for the cloning technology and industrialization of transgenic breeding.
