Abstract


The swine streptococcus (Streptococcus suis, SS) is against the modern pig industry, an important Zoonosis pathogenic bacteria serotype antigen complex structure, can present different clinical characteristics, such as sepsis, meningitis , endocarditis, pneumonia, arthritis, often combined with other diseases, infections, high morbidity and mortality. Of Streptococcus suis Occurrence, the pathogen characteristics as well as the prevention and control measures is not very clear, simply relying on drugs can not effectively control the occurrence and spread of the disease. In 2005-2009, the number of pigs reared in large-scale pig farms in the Yellow River mouth area varying degrees of Streptococcus suis occurred. Studying the test from the etiology, cultured Streptococcus suis strains in the separation of the Yellow River mouth area, identified on the basis of the establishment of the PCR method for virulence gene detection, in order to provide a theoretical basis for the disease prevention and control of immune. This topic is divided into two parts of the study. 1 Yellow River mouth area of ??the separation and identification of pathogenic Streptococcus suis resistance Survey collection 2005 -2009 region-scale pig farms in the Yellow River Mouth suspected Streptococcus suis sick pigs, according to clinical diagnosis, pathology observation, combined with pathogen isolation culture and identification, diagnosis of Streptococcus suis. By Gram staining, biochemical tests and susceptibility testing of clinical isolates of Streptococcus Suis epidemiological preliminary study. 15 isolated from the region, the incidence of swine Streptococcus Suis, morphology, staining, culture characteristics and physical and chemical characteristics to meet the the Streptococcus suis characteristics, susceptibility test results show that the Yellow River mouth area Streptococcus suis only ampicillin, amikacin, cefotaxime, cefazolin sensitive to other conventional drugs produce different degrees of resistance. According to the Streptococcus suis two main virulence factor gene Sly and the MRP gene sequences, the double PCR analysis of the two major virulence factor of Streptococcus suis, design and synthesis of the two pairs of specific primers optimized through the system and conditions, to establish a double PCR detection methods, saved 15 clear background swine Streptococcus strains were detected. The results show that the the Sly detection rate was 4/15, MRP detection rate of 2/15, positive and negative controls are set up. The analysis showed that the the double PCR detection method, good specificity and sensitivity, molecular epidemiological investigation can be used for the rapid diagnosis and Streptococcus suis virulence factors.
