Abstract


The test for the optimization of sheep oocytes in vitro culture system , to provide experimental basis as the research object , sheep , sheep ovaries were observed using histological techniques based on the comparison of the extraction method and cutting method to obtain the eggs in the ovarian surface follicles the effect of mother cells , and in vitro maturation studies , histochemical techniques at the same time , to observe the general morphology of different developmental stages of oocyte lipid content . The results showed that : the sheep ovary oval, the number of primordial follicles and primary follicles scarce than other livestock the the three follicular phase can be seen the formation of cumulus . Compared with suction wears cutting method available number of oocytes collected and cultured diffusion rate and maturity of oocytes was significantly higher than the suction method ( P lt; 0.05 ) , and lower than the bare egg rate cut method aspiration . In vitro oocyte maturation process , can significantly improve the oocyte maturation rate add 0.095mg/mL cysteine ??. Less lipid content within the oocyte Oh development of oocytes, lipid droplets distribution density of uniform , dense , 8h lipid droplets slightly decreased lipid droplets larger , 24h , 30h blastomeres lipid droplets of different sizes, distribution uneven density .
