Agronomic and Genetic Evaluation of a Bud Sport of ’Fuji’ Apple

Abstract


Spontaneous mutation often occurs in apple,especially in Delicious Apples.Some bud mutation can be directly used for breeding.Here we analyzed the quality of one ’Fuji’ redder skin color bud sport and molecular characterization were performed between the mutant and the progenitor(the source of the tree where mutation happens) by molecular markers,including AFLP(Amplified fragment length polymorphism),MSAP(methylation - sensitive amplified polymorphism)and SSAP(Sequence-specific amplification polymorphism).The main results are as follows:1.The absorbance of anthocyanin extracted from the skin of bagged ’Fuji’ bud sport mature fruit is 118.62U per 100 cm~2,remarkably higher than that of contrast(88.92 U/100 cm~2),this is in accordance with mutation phenotype that bud sport apple has a bright redder skin color if bagged.The flavor of ’Fuji’ bud sport mature fruit is rich.The contents of fructose(59.6 mg/g) and glucose(10.9 mg/g) in ’Fuji’ bud sport non-bagged mature fruit are lower than thoes of contrast(fructose72.1 mg/g,glucose14.6 mg/g),then the content of sucrose(23.4 mg/g) is highter than that of contrast(18.5 mg/g),and the content of malic acid(1.7 mg/g) is lower than contrast(2.5 mg/g),so the quality of the ’Fuji’ bud sport non-bagged mature fruit is close to contrast.The contents of fructose,sucrose and glucose in ’Fuji’ bud sport bagged mature fruit are all higher than those of contrast,the content of malic acid is highter slightly than that of contrast.The results showed that the mutation of skin color of ’Fuji’ bud sport did not change the furits’ inherent superiot quality,the quality is even better.2.The ploidy of ’Fuji’ bud sport and contrast was analysed by the flow cytometry. The results indicated that they were all diploids.3.Using 269 pairs EcoRⅠ/MseⅠprimers in AFLP analysis,18000 bands were amplified,of which unfortunately there was no polymorphic band.And in MSAP analysis 4704 bands were amplified by using 56 pairs EcoRⅠ/HpaⅡ- MspⅠprimer and 56 pairs MseⅠ/PstⅠprimers.One polymorphic band was found between ’Fuji’ bud sport and contrast.We recovered the fragment and sequenced it.The sequence information indicated that it is of 120 bp in length,and had 42%homology with the phosphoglycan-beta-1,3-galactosyltransferase gene according to the Genbank BLAST analysis.Moreover,SSAP analysis with 32 pairs EcoRⅠ/LTR primer and 32 pairs MseⅠ/LTR primer produced 3264 bands,of which one band showed polymorphism between the bud mutant and the wild type.After recovering and sequencing,we obtained a DNA fragment of 376bp in length;and BLAST against GenBank database revealed that it is 90%homologous to the Promoter of chalcone synthase(CHS) gene, which is a key enzyme of the anthocyanin biosynthesis.The interesting data is consistent with our previous observation that the major accumulated pigment in skin of the apple mutant is anthocyanin.All the results indicated the molecular polymorphism between the ’Fuji’ bud mutant and the wild type can be detected on DNA level,and the cloning of the polymorphic loci paved a basis for further research on the molecular mechanism of bud mutation in the red-skin apple.The results indicated the ’Fuji’ bud sport was new germplasm and could be applied to the industry as elite cultivar.
