Abstract

Objective: 125 the I particle inter-organizational implantation is a low dose rate continuous ionizing radiation treatment has a better therapeutic effect. It as a stand-alone treatment, in a foreign country has been widely used in the treatment of prostate cancer and other solid tumors, and to be able to reach a satisfactory long-term local control rates. But 125 I seed implantation of radiation injury to normal tissues, organs not yet determined data. The study by the different doses 125 I particles implanted in rabbit trachea, esophagus, arteries adjacent to observe the different radiation dose, the pathological changes of the different time points, the evaluation 125 I seeds implanted in the structure of the tissues and organs of the rabbit, and its security, provide the basis for rational clinical application. Method: the general level of 120 male rabbits the rabbit age around June, purchased from Zhengzhou University School of Medicine Animal Department. According to the experimental design were randomly divided into four groups, namely the control group (12), trachea (36), the esophageal group (36), arterial group (36); each experimental group again into 0.5mCi group, 0.8mCi group 1.5mCi group three subgroups, each of 7 days, 14 days, 30 days, 60 days, four time points drawn. The general animal before and after implantation of the observed changes in the situation, according to the preoperative and the four time points extracted peripheral blood leukocyte, red blood cells, hemoglobin changes, drawn after the trachea, esophagus, arteries with 4% paraformaldehyde fixed, paraffin-embedded the conventional slice HE staining sections by two physician pathology (based on the principle of double-blind) in light microscopy, and injury severity score (Sun Jian and other the on 103 Pd stent continues of radioactive bile duct injury classification method improvements); conventional slice with immunohistochemistry method for detection of Bax protein expression, detection methods for the quantitative analysis of positive pressure integral optical density. All values ??are expressed as mean ± standard deviation ((?) ± s), homogeneity of variance by Levene analysis, analysis of variance (SNK-q) inspection SPSS11.0 statistical software processing is completed, P < 0.05 was considered statistically significant. Results: 1 general: animals implanted particles incision healed well, no infection; were normal diet, anorexia, nausea, vomiting, keeping to the drawn period, increases in body weight, blood cells are maintained at a relatively stable level not big fluctuations (P> 0.05). 2. Conventional pathological observation: two physician pathology in light microscope biopsy, and injury severity scoring. Experimental group organizations have different levels of damage, and particle radiation dose, and injury severity increased with radiation dose (P <0.05), the experimental group damage is reversible, were restored to normal in 60 days, the difference with the normal control group was not statistically significant (P> 0.05). Damage repair and particle radiation dose on, 1.5mCi group of slow recovery in 0.8mCi group, and 0.5mCi of group, the difference was statistically significant (P <0.05). The degree of damage to tissue differences esophagus trachea> artery. 3. Bax protein expression: normal tissue Bax protein expression is negative, the experimental group at 7 days after Bax protein expression in various organizations increased (P <0.05), to the day 14 Table peaked (P <0.01), and continued to 30 days and subsequently decreased, the organization also entered the recovery phase. To 60 days experimental group expression in comparison with the control group, the difference was not statistically significant (P> 0.05), close to the normal structure of the organizations. Meanwhile, the results showed that the the 1.5mCi group at each time point in the 60 days prior to Bax expression were higher than those the 0.5mCi group, and 0.8mci of group statistically difference was statistically significant (P <0.05). 4. The study found that the tissue damage epithelium, connective tissue, myometrial only a small amount of damage cells. Conclusion: of a radioactive 125 the I particles of normal trachea, esophagus, arterial tissue damage, and particles radiation dose, but the damage is reversible, normal tissue in a certain range of radiation dose self-repair. 2 normal tissue tolerance of γ-ray is a certain threshold radiation dose 0.8mCi following applications are safe 1.5mCi radiation dose of long-term use may induce increased apoptosis, resulting in irreversible cell damage. 3 different organizations 125 the degree of tolerance of the I particles: the mucosal tissue of the artery> trachea esophagus; myometrial tissue connective tissue>.
