Objective To study the apoptosis rate and the activity of ROS and the inhibitory action of superoxide dismutase (SOD) to apoptosis rate and the activity of ROS of lens epithelial cells of rabbit induced ultraviolet( UV) radiation.Methods 130 lens of rabbit were randomly divided into A group(contrast group)B group(UV irradiated group)and C group (UV+SOD) in MEM cultural medium.Cultured rabbit lenses were exposed to UV radiation in MEM cultural medium without or with superoxide dismutase (SOD) except A group(contrast group). Lenses epithelium cells were harvested to estimate the intracellular reactive oxygen levels by staining with 2’,7’-dichlorofluorescein diacetate(DCFH-DA) by flow cytometry.Annexin V/PI two-color flow cytometry was used to detect the apoptosis.Results The observations revealed, UV radiation could apparently induce lens epithelial cell apoptosis in vitro, the apoptosis rate increased to (36.0621±2.5224) % after exposure to UV, which is also the time a higher level of intracellular ROS increased to (98.4502±0.5606) % detected in the irradiated group and SOD could inhibit lens opacification, apoptosis and ROS activation. The apoptosis rate decreased to (12.9384±0.7249) % ,( P < 0.05), the level of intracellular ROS decreased to (89.5625±0.4502) %,(P<0.05).Conclusion Lens epithelium cell are affected by UV radiation. These data indicate that antioxidants SOD, possibly through inhibit of the activity of ROS, can prevent lens opacification and apoptosis of lens epithelial cells.
