Abstract

Wolfberry (Lycium Barbarum. L) Solanaceae (Solanaceae) plant genus of Chinese wolfberry (Lycium), Ningxia \health care products. Wolfberry is a perennial deciduous shrub, is also the major economies of the Ningxia region one of the trees. Recent years, with further development, the cultivation area of ??the wolfberry medicinal value expanding conventional breeding methods can not meet the needs of modern agriculture wolfberry improved varieties and to cultivate the wolfberry new varieties method rarely. Haploid breeding methods anther culture techniques haploid plants homozygous diploid after after doubling, in breeding very early stable separation offspring, shorten the breeding period, to simplify breeding program. Wolfberry anther culture predecessors although research, but only remain in the theoretical discussion on no real wolfberry anther culture technique for production practice. This study are several factors that affect wolfberry anther culture (basal medium, hormones, activated carbon, temperature, mannitol pretreatment) system, designed to be regenerated plants and transplanting optimization wolfberry anther culture conditions, establishment of a technical system for wolfberry anther culture, wolfberry breeding to improve efficiency, accelerate the process of the wolfberry breeding breeding, to provide a theoretical basis for the the wolfberry genetic improvement and genetic mechanisms, so as to promote the rapid development of the Chinese wolfberry industry. Test to Ningqi No. 1, cannabis leaf, 0207, Meng Qi No. 1 material, L32 (2 × 45) orthogonal design anther browning affect wolfberry, the rate of callus induction and somatic embryo of rate factors were studied. The test results are as follows: 1, of different genotypes material anther culture there are significant differences in the four genotypes inoculated browning callus and embryoid induction rate difference between the larger Ningqi 1 is easier induction genotypes. 2, two basic medium study found: MS than B5 the more suitable Ningqi 1 and cannabis leaves anther culture; 0207 and Meng Qi, a higher rate of induction on B5. 3, plant growth regulators play an important role in the Wolfberry anther culture using different concentrations of 2,4-D, KT ratio, 1.0mg/L2 ,4-D and 1.0mg/LKT the ratio suitable for the formation of callus. 4, the approach of the three temperatures, 4 ° C bud low temperature pretreatment temperature by 32 ℃ cultured for one day and then transferred to 25 ° C dark culture most suitable for anther culture Ningqi 1. 5 medium 0.8g / L of activated carbon can effectively reduce the degree of browning of anthers, and can increase the incidence of embryoid; However, the addition of activated carbon was reduced callus induction rate, and activated carbon concentration more The high induction rate is lower. Before inoculation of anthers the mannitol pretreatment can improve the rate of callus induction, the mannitol concentration 0.3mol / L is the best. 7, orthogonal experiment results show that: the wolfberry anther culture of genotype and plant growth regulator is very dependent; activated carbon significantly affected anther browning rates, which affect the primary and secondary order: activated carbon gt; mannitol gt; basic medium gt; thermal shock treatment gt; plant growth regulators; basic medium and mannitol callus reached a significant level, the order was: plant growth regulators gt; basic medium gt; mannitol gt; activated carbon gt; heat shock treatment; activated carbon of embryoid incidence difference is extremely significant, basic medium heat shock treatment, the the embryoid incidence reached significant level, with the order of: Plants growth regulator gt; activated carbon gt; basic medium gt; heat shock treatment gt; mannitol.
